Objective:Different subgroups of helper lymphocyte T cell play different roles across the development of atherosclerosis.To investigate the relationship between helper lymphocyte T cell subgroups change and AMI as well as the development of coronary artery disease.We analysis the different expression of helper lymphocyte T cell subgroups Th1,Th2,Th17 and cytokines such as IFN-γ,IL-4,IL-17A within AMI patients ,UA patients and the patients with chest pain but normally coronary arteriongraphy.Methods:1) Density gradient centrifugation was employed to isolate mononuclearcell from AMI patients(n=15),UA patients(n=15) and patients with chest pain and coronary arteriongraphy are normal(n=15).2)Flow cytometric analysis was applied to detect the numbers of Th1,Th2 and Th17. Mononuclearcell was incubated for 4 hours with the condition of 37°C,5%CO2 and Brefield A.Then staining the Mononuclearcell and analysis it by Flow cytometric.3)The plasma levels of IFN-γ, IL-4 and IL-17A were analysed by ELISA.Results:1) control group,UA group and AMI group have no significant differences of risk fator,such as age,sex, blood fat , blood glucose ,hypertention and smoking,etc.2) Flow cytometric analysis was applied to detect the numbers of Th17,Th1 and Th2.①The percentage of Th17,Th1,Th2 in peripheral blood of AMI group,UA group and control group.The percentage of Th17 in peripheral blood of AMI group(0.35±0.05%) is higher than that in UA group(0.25±0.03%) and control group(0.12±0.01%)(P<0.01), and UA group is higher than control group(P<0.05);The percentage of Th1 in peripheral blood of AMI group is higher than that in UA group(0.33±0.05%) and control group(0.16±0.02%)(P<0.05), and UA group is higher than control group(P<0.01);The percentage of Th2 in peripheral blood of AMI group (0.53±0.11%) is higher than that in UA group(0.17±0.02%) and control group(0.30±0.05%)(P<0.01), control group is higher than UA group(P<0.01).②The cell numbers of Th17 are greater in occlusion coronary artery(0.58±0.14%) compared with peripheral blood(0.35±0.05%) of AMI group(P<0.05). However, the levels of Th17 in coronary artery(0.31±0.03%) have no significant difference with peripheral blood(0.25±0.03%) of UA group(P>0.05),the levels of Th17 in coronary artery(0.14±0.01%) have no significant difference with peripheral blood(0.12±0.01%) of control group(P>0.05);The cell numbers of Th1 are greater in occlusion coronary artery(0.76±0.13%) compared with peripheral blood(0.52±0.12%) of AMI group(P<0.01). However, the levels of Th1 in coronary artery(0.32±0.05%) have no significant difference with peripheral blood(0.33±0.05%) of UA group(P>0.05),the levels of Th1 in coronary artery(0.16±0.01%) have no significant difference with peripheral blood(0.16±0.02%) of control group(P>0.05). The cell numbers of Th2 are greater in occlusion coronary artery (0.84±0.20%) compared with peripheral blood(0.53±0.11%) of AMI group(P<0.01). However, the levels of Th2 in coronary artery(0.19±0.03%) have no significant difference with peripheral blood(0.17±0.02%) of UA group(P>0.05),the levels of Th2 in coronary artery(0.30±0.03%) have no significant difference with peripheral blood(0.30±0.05%) of control group(P>0.05).3) The plasma levels of IFN-γ, IL-4 and IL-17A .were analysed by ELISA.①The plasma levels of IL-17A in peripheral blood of AMI group (250.04±47.58pg/ml) is higher than that in UA group(130.5±16.97 pg/ml) and control group(59.86±5.70 pg/ml)(P<0.05), and UA group is higher than control group(P<0.05);The plasma levels of IFN-γin peripheral blood of AMI group(73.3±9.87 pg/ml) is higher than that in UA group(33.26±6.15 pg/ml) and control group(15.03±0.51 pg/ml)(P<0.05), and UA group is higher than control group(P<0.05);The plasma levels of IL-4 in peripheral blood of AMI group(8.93±2.33 pg/ml) has no significant difference with control group(12.63±1.24 pg/ml)(P>0.05).②The plasma levels of IL-17A are greater in occlusion coronary artery(529.91±57.17 pg/ml) compared with that in peripheral blood (250.04±47.58 pg/ml) of AMI group, P<0.01. However, the levels of IL-17A in coronary artery(247.47±64.39 pg/ml) have no significant difference with peripheral blood(130.5±16.97 pg/ml) of UA group(P>0.05),the levels of IL-17A in coronary artery(80.66±10.98 pg/ml) have no significant difference with peripheral blood(59.86±5.70 pg/ml) of control group(P>0.05).The plasma levels of IFN-γare greater in occlusion coronary artery(102.08±19.42 pg/ml) compared with peripheral blood(73.3±9.87 pg/ml) of AMI group, P < 0.05. However, the levels of IFN-γin coronary artery(25.48±7.06 pg/ml) have no significant difference with peripheral blood(33.26±6.00 pg/ml) of UA group(P>0.05),the levels of IFN-γin coronary artery(16.63±1.18 pg/ml) have no significant difference with peripheral blood(15.03±0.51 pg/ml) of control group(P>0.05).The plasma levels of IL-4 are greater in occlusion coronary artery(17.25±4.5 pg/ml) compared with that in peripheral blood(8.93±2.33 pg/ml) of AMI group, P<0.01. However, the levels of IL-4 in coronary artery(12.28±1.34 pg/ml) have no significant difference with peripheral blood(12.63±1.24 pg/ml) of control group(P>0.05).Conclusion: The study found that the number of Th1,Th17 and cytokines of IFN-γ,IL-17A increased following the aggrevation of artherosclerosis. The numbers of Th2 in AMI patients is higher than that in Control group, however, UA group is lower than control group.The study showed that Th17 was activated with the development of artherosclerosis and AMI, and the immune response is higher in local occlusion coronary artery than in peripheral blood.So we can draw the conclusion that Th1,Th17 can aggrevate the unstalility of artherosclerosis.This provide a new method of regulate and stabilize artherosclerosis plaque.
