Ovarian cancer is a serious threaten to the lives of women.Because it is hard to diagnose at a ealy stage,many women lost the best opportunity of medical treatment.And the mortality rate is high.So it is really necessary to find a better way to diagnose the ovarian cancer.The laboratory have found M/Z5908 and M/Z5640 high expressed in the serum of women with ovarian cancer by SELDI-TOF-MS technology.Further experiments identified M/Z5908 protein as Amyloid beta A4 protein(APP),which is confirmed by detecting its content in serum of patients with ovarian cancer.And M/Z5640 has not been identified.To further clarify the diagnostic value of the Amyloid beta A4 protein in ovarian cancer,this study detect the expression ofβ-amyloid A4 protein in cases of ovarian cancer,benign ovarian tumor and normal ovarian tissue by real-time fluorescence quantitative PCR,and revealed the relationship between the its expression and clinical facter.Many reported has found thymopoietin protein high.expressed in cancer,whose M/Z is reportde M/Z5638.In order to identify whether M/Z5640 is the thymopoietin protein,this study detected the content of thymopoietin in cases of ovarian cancer,ovarian benign tumor and serum of health women by ELISA and explore the relationship between its expression and its clinical facter. Objective To explore the relationship between the mRNA expression ofβ-amyloid A4 protein in ovarian cancer and its clinical-pathology facter.Methods The total RNA of the ovarian tissue of the patients with ovarian tumor was extracted by one-step Trizol extraction method.The expression ofβ-amyloid A4 protein was detected by real-time fluorescence quantitative PCR in 49 cases of ovarian cancer and 20 cases of benign ovarian tumor and 16 cases of normal ovarian tissue.Results The APP expression of the ovarian cancer compared with benign ovarian tumor and normal ovarian tissue is at a significantly higher lever(P value<0.05);The APP expression of the ovarian granulose cell tumor is significantly higher than that in epithelial ovarian cancer(P value<0.05);The was not significant correlation between APP expression and the survival rate of patients with ovarian cancer,and Cox analysised also suggested that APP expression is not an independent judgment of the prognosis of patients with malignant ovarian tumor.Conclusion The APP expression of the ovarian cancer is significantly higher than that of benign ovarian lesions,and it is particularly high expressed in ovarian granulose cell tumor,suggesting that its high expression may be related with its histology and malignant type and development of canciroma. Objective To detected the content of thymopoietin in serum of patients with ovarian cancer and explore the relationship between its expression and its clinical facter.Method The expression of thymopoietin in serum of 50 cases of ovarian cancer,34 cases of ovarian benign tumors and 40 normal healthy women was detected by ELISA.Results Levels of expression of thymopoietin in serum of patients with ovarian cancer,benign ovarian tumors and normal ones is not statistically significant differences(P<0.05);Differences of thymopoietin expression in ovarian cancer with different histological grade,clinical stage,metastasis and pathological types were not statistically significant.ROC curves show malignant ovarian tumors can not be distinguished by expression of thymopoietin.Conclusion There was no obvious increase of thymopoietin expression in serum of patients with ovarian cancer.Thymopoietin can not be used as diagnostic indicators of ovarian cancer.
