Objective To investigate the changes of IL-17 level in peripheral blood mononuclear cells(PBMCs) with dilated cardiomyopathy(DCM),and the relation of IL-17 level in PBMCs with DCM disease severity.Explore the role of IL-17 in human DCM.Methods 30 cases of patients with DCM,26 cases non-DCM patients and 30 cases normal adults were studied.IL-17 protein level in plasma,PBMCs culture supernatants,and Phytohemagglutinin(PHA) stimulated PBMCs culture supernatants were measured with Enzyme-linked Immunosorbent Assay (ELISA).IL-17 mRNA expression in PBMCs was detected by Reverse Transcription-Polymerase Chain Reaction(RT-PCR).Results(1) IL-17 protein level in PHA stimulated PBMCs culture supernatants,but not in plasma or in PBMCs culture supernatants,was significantly elevated in DCM patients(193.58±19.65) compared with non-DCM patients(62.03±6.75) and normal adults(59.52±11.28).There was a significant difference(P<0.01).(2) The expression of IL-17 mRNA in the PBMCs of no culture and medium culture was not detected in DCM patients, non-DCM patients and normal adults.However,IL-17 mRNA expression was apparent and could be seen in PBMCs stimulated with PHA.Compared with non-DCM patients(0.57±0.22) and normal adults(0.51±0.14),IL-17 mRNA expression was significantly higher in DCM patients(0.84±0.27) in PBMCs stimulated with PHA(P<0.05).(3) The IL-17 protein level in cultured supernatants and the IL-17 mRNA expression in the PBMCs were strongly positively correlated with heart function(r=0.76,P<0.01;r=0.81,P<0.01) of DCM patients,and were negatively correlated with ejection fraction(r=-0.77, P<0.01;r=-0.69,P<0.01) of DCM patients.Conclusions(1) IL-17 protein level or mRNA expression in the PBMCs was significantly elevated in DCM patients.(2) The change IL-17 protein level or mRNA expression was strongly correlated with heart function of DCM patients.(3) IL-17 maybe involve in the pathogenesis of DCM,and may play an important role in the pathogenesis process of DCM...
