Objective To study the mechanism of mangiferin-induced apoptosis in HL-60 keukemic cells.Methods The standard HL-60 cells were divided into control and experimental groups,expression levels of apoptosis-related genes incluing bcl-2 and survivin were determined by RT-PCRResult 1. 20-250μumol/L mangiferin down-regulated the expression levels of bcl-2gene in HL-60 cells after treatment with mangiferin for 24 hours,and the proportion of wich with 250μmol/l mangiferin was significantly lower than that occurring in untreatd cells(0.77±0.03vs0.49±0.03,p<0.01).also the proportion of cells after treatment with 250u mol/l mangiferin for 48 hour was lower than that treated cells for 24 hours(0.49±0.03vs0.35±0.04 p<0.01).2. 20-250μmol/L mangiferin down-regulated the expression levels of survivin gene in HL-60 cells after treatment with mangiferin for 24 hours,and the proportion of wich with 250μmol/l mangiferin was significantly lower than that occurring in untreatd cells(0.88±0.03vs0.59±0.02,p<0.01).and the proportion of cells after treatment with 80μmol/l mangiferin for 48 hour was lower than that treated cells for 24 hours(0.69±0.02vs0.60±0.02 p<0.01).Conclusion The mechanism of mangiferin-induced apoptosis in HL-60 keukemic cells might be in down-regulated the expression levels of bcl-2 and survivin genes in HL-60 cells.
