Objective:To investigate systematically the existence of bactericidal/ permeability increasing protein (BPI),β-Defensins (DF)-1 and platelet phospholipase A2 mRNA in normal rat organs and tissues,for providing important information in clinical appli- cation。To investigate the effects of cyclophosphamide (CTX) on anti-bacterial protein mRNA in rats, for understanding the immune inhibition mechanism of CTX。Methods: Total RNA was extracted by Trizol from the rats organs and tissures that each weighted 50-100mg, RT-PCR was performed to amplify the cDNA of BPI,β-Defensins-1 and platelet phospholipase A2 ,the products were conducted by electrophoresis and followed by semi-quantity analysis; Cyclophosphamide(40mg/kg) was injected into rats'abdominal cavity for five days , while in the control group , normal saline was injected。after finished ,the cintents of BPI,β-DF-1 and platelet phospholipase A2 were detected from the rats organs and tissures。ResuLt:(1)BPI mRNA were found in kidney,ovary,testis,liver,small intestine,large intestine,thymus,spleen and neutrophils of 21 various organs and tissues。Among them ,BPI mRNA content was the richest in testis,relatively rich in liver and large intestine。(2)β-Defensins-1 mRNA were found in trachea,cardia,kidney,uterus,ovary,testis,liver,stomach,small intestine,thymus,brain and skin of 21 various organs and tissues。Among them ,β-Defensins-1 mRNA content was relatively rich in trachea,cardia,kidney,stomach,ovary and skin。DNA sequencing not only proved the PCR product wasβ-Defensins-1 but also proved it was the same as general acceptedβ-Defensins-1 structure。(3) BPI,β-Defensins-1 and platelet phospholipase A2 mRNA were found in testis,epididymis and ovary of 7 various genital organs and tissues。Among them ,BPI mRNA of testis andβ-Defensins-1 mRNA of epididymis and platelet phospholipase A2 mRNA of ovsry content were rather aboundance。Moreover,β-Defensins-1 and platelet phospholipase A2 mRNA were also found in prostatic gland,uterus and vsgina。(4)In normal control group(NS),BPI was all found intestis,epididymis,thymus,liver,small intestine and large intestine ;in CTX group,BPI was not found in thymus and liver,and its content was found in other's tissue but lower than NS group。In NS group,β-Defensins-1 was found in testis,epididymis,thymus,liver and small intestine,the richest in epididymis;but in CTX group,it was not found in intestis,thymus,liver and large intestine ,only in epididymis and small intestine,and the content was lower than NS group。In NS group,platelet phospholipase A2 was all found in testis,epididymis,thymus,liver,small intestine and large intestine ;in CTX group,it was not found in intestis and thymus ,only in epididymis,liver,small intestine and large intestine,and its content was also lower than NS group。Conclusions:(1)BPI mRNA andβ-Defensins-1 present in many organs and tissues of rat and it indicates that the role of BPI andβ-Defensins-1in controlling bacteria infection is relatively extensive。(2)BPI,β-Defensins-1 and platelet phospholipase A2 mRNA present in many organs and tissues of rat genital system ,and it indicates that anti- bactericidal protein take important part in controlling bacteria infection。(3)CTX inhibit the expression of anti-bacterial protein,that indicate the immune inhibition of CTX is implemented not only by inhibiting bone marrow but also by inhibiting anti- bacterial protein。...
