Objective To evaluate the effects of local application of basic fibroblast growth factor(bFGF) on angiogenesis in ischemic limbs of rats in different age groups.Methods 40 Wistar rats were equally divided into two groups based on the age: group A(4-month-old) and group B(16-month-old).Group A was evenly subdivided into groups A1 and A2,and Group B into groups B1 and B2.The left femoral artery of all the rats were cut and ligated,an ischemic model completed.Intramuscular injection of recombinant bovine basic fibroblast growth factor(rb-bFGF) was given to groups A1 and B1 in ischemic zone,the equality of normal saline to groups A2 and B2.Four weeks postoperatively,the adductor tissue was sent for histology,the blood-vessel density was calculated by image analysis,and positive VEGF by immunohistochemistry.Results The member of capillaries and positive VEGF was more in group A1 than that in A2,more in group B1 than B2(t=3.30-3.50,P<0.05),but the difference between other groups was not significant(P>0.05).Conclusion Recombinant bovine bFGF can stimulate angiogenesis and improve the ischemic limb of rat;which is not associated with their age.
