Objective:To discuss the clinical application value of the expressions and serum levels of neuron-specific enolase(NSE) and S-100 proteins and the apparent dispersion coefficient (ADC) value in magnetic resonance diffusion weighted imaging(MRI-DWI) in diagnosing cerebral infarction and monitoring the development of cerebral infarction by analyzing the changes of the expressions and serum levels of NSE and S-100 proteins and the ADC value in MRI-DWI after middle cerebral artery occlusion and reperfusion (MCAO/R) in rabbits.Methods:Fifty-eight successful MCAO/R rabbit models were divided into permanent ischemia group(A group,n=30)which were subdivided into ischemic 1h,3h,6h,12h,24h,48h groups(n =5 each grpup) and reperfusion group after 1h cerebral ischemia(B group,n=28)which were subdivided into 2h(n=5),5h(n=5), 11h(n=4),23h(n=5),47h(n=4) groups.Another 10 rabbits were regarded as ischemic control group.All rabbit brains were cut out to make histologic section immediate after each time point MRI examination,then measured the ADC value.The expressions of NSE and S-100 proteins in the brain tissue were determined with immunochemical assay, and the serum levels of NSE and S-100 proteins were measured with ezyme linked immunosorbent assay(ELISA).Results:The expressions of NSE and S-100 proteins increased markedly following permanent ischemic 1h,and reached a peak at permanent ischemic 12h and ischemic reperfusion 11h.After that,the expressions of NSE and S-100 proteins was gradually descent,increase again.The expressions of NSE and S-100 proteins in reperfusion groups were weaker than that in permanent ischemic groups,and increased again at reperfusion 23h~47h.The serum levels of NSE and S-100 proteins increased markedly at ischemic 6h and reperfusion 5h,and reached a peak at permanent ischemic 24h and ischemic reperfusion 23h.and decreased at permanent ischemic 48h and ischemic reperfusion 47h.The changes of the serum levels of NSE and S-100 proteins were delayed 5h than that in brain tissue expression in permanent ischemic groups and permanent ischemic groups.The average ADC value decrease at frist and then increase gradually at different time points.There was significant singnals with the decrease of ADC value at ischemic 1h point in permanent ischemia groups,and the low peak is at permanent ischemic 6h and ischemic reperfusion 5h,and decreased again at reperfusion 23h~47h,which was corresponded with the expression increases of NSE and S-100 proteins.Conclusions:1.The expressions of NSE and S-100 proteins in the brain tissue was impressionable and specific in discovering cerebral infarction.The expressions of NSE and S-100 proteins increased again at reperfusion 23h~47h,which indicated reperfusion injury.2.MRI-DWI and the ADC value were highly impressionable and specific in discovering cerebral infarction and reperfusion injury,and even outweigh the expressions of NSE and S-100 proteins in the brain tissue.3.The serum levels of NSE and S-100 proteins had significant changes following cerebral infarction,and the time was markedly later than that of the expressions of NSE and S-100 proteins.This study didn't reflect the reperfusion injury on serology.Moreover,the increases of the expression of NSE in reperfusion groups was higher than that in ischemic groups,which possiblely mislead to estimationing the degree of brain damage.
