| Purpose: To observe the effect of IFNα-2b combined with BCG on the Wistar rats model of bladder cancer and discuss the mechanism of the therapeusis.Methods: 50 female Wistar rats, 6～7 weeks old, were irrigated MNU into their bladders to induce the bladder cancer. After two weeks executed 10 rats randomly and observed the growth of bladder cancer. Other 40 rats were separated into 4 groups randomly, which is PBS group(A group),Interferon group(B group),BCG group(C group),drug combination(D group), They were treated by way of irrigation of bladder with either 0.2ml PBS, 0.1ml IFN, 0.1ml BCG, 0.1ml IFN+0.1ml BCG respectively every week(10th, 11th and 12th week) for a total of three times. In the thirteenth week, executed all rats, observed the growth of bladder cancer. Detected the number of CD3,CD4 and CD8 cells around the tumor tissue through immunohistochemistry. Detected the apoptosis of cancer cells through TUNEL.Results: The weight of bladder in D group was lower than other groups (P<0.05); the weight of bladder in C group was lower than A and B groups (P<0.05). The percentages of the number of CD3,CD4 cells and CD4/CD8 around tumor tissue in D group were higher than other groups (P<0.05); the percentages of the number of CD3,CD4 cells and CD4/CD8 around tumor tissue in C group were higher than A and B groups (P<0.05). The apoptosis rate of bladder cancer in D group was higher than other groups (P<0.05); the apoptosis rate of bladder cancer in C group was higher than A and B groups (P<0.05).Conclusion: 1. Drug combination can increase the number of immune cells around cancer, stimulate autoimmunity, and furthermore participate the immune therapy of bladder cancer. 2. The growth inhibition and apoptosis of BCG, which is enhanced by IFNα-2b, is maybe the mechanism of IFNα-2b combined with BCG in the treatment of bladder cancer. 3. The irrigation of bladder with BCG has some effect in the treatment of bladder cancer; while the effect of the irrigation of bladder with IFN was not well.
