Objective:To study the influence of Angâ…¡and Ang-(1-7)on ABCA1 mRNA,protein and cholesterol efflux in THP-1 derived macrophages. Methods:Cultured THP-1cells were induced to differentiate into macrophages by PMA for 48hours. The macrophages were randomly divided into control group, Angâ…¡groups, Ang-(1-7) group, Ang-(1-7) + Angâ…¡group, and Ang-(1-7) + Angâ…¡+A-779 group. The expression of ABCA1 mRNA and protein was measured by RT-PCR and western-blot respectively and observed macrophage cholesterol efflux. Results:Compared with the control group, Angâ…¡reduced the expression of the ABCA1mRNA and protein ,inhibited the cholesterol efflux(P<0.05).Ang-(1-7) dose-dependently opposed the reduction of ABCA1 induced by Angâ…¡,and promoted the cholesterol efflux(P<0.05),and A-779 did not affect the expression of ABCA1 and cholesterol efflux. (P>0.05) Conclusion: The effects of Angâ…¡on the formation of foam cells and atherosclerosis may be correlated to the down-regulation of ABCA1. Ang-(1-7)concentration-dependently attenuate the reduction of ABCA1 induced by Angâ…¡in THP-1 derived macrophages through its specfic receptor Mas.
