Background: The glycuresis complication Diabetic Nephropathy (DN) is one of main dead cause of glycuresis patient, the life the glycuresis patient being threatening gravely.Transforming growth factorβ1 (TGF-β1) and Intercellular adhesion molecule-1 (ICAM-1) playing main role in occurrence and development in DN. Adiponectin (ADPN) is to find having various Biology activity in recent years, but the role of ADPN is still ambiguous in kidney. Therefore, The experiment makes use of extracorporeal culture rat mesangial cells (RMC) to study the effect of TGF-β1 and ICAM-1, to discuss the effect of ADPN in DN.Objective: To investigate the effect of high glucose on the expression of Transforming growth factorβ1 (TGF-β1) and Intercellular adhesion molecule-1 (ICAM-1) in rat mesangial cells (RMC), and to further study the intervention of adiponectin (ADPN). To illuminate the potential protection of ADPN on the kidney.Methods:①The RMC were allocated into 3 groups: normal glucose group (5.5 mmol/L glucose,NG) , high glucose group (25.0mmol/L glucose, HG) , HG +ADPN group(25.0mmol/L glucose+2.5μg/ml adiponectin:), After RMC was cultured with upper three different kinds of environments for 24h,48h,96h ,the proliferation activity of RMC was observed by MTT .②The RMC were allocated into 3 groups: normal glucose group (5.5mmol/L glucose,NG), high glucose group(25.0mmol/Lglucose,HG), HG+ADPN group( 25.0mmol/L glucose+different concentrations of adiponectin: 2.5, 5.0, 7.5, 10.0, 15.0, 20.0μg/ml). After RMC was cultured with upper three different kinds of environments for 48 hours, the expression of TGF-β1 and ICAM-1 was measured by ABC-ELISA.Results: Compared with NG group, proliferation activity of RMC was inhibited in high glucose group. And compared with HG group , proliferation activity of RMC increased time-dependently in low concentration adiponectin group(2.5μg/ml), The expression of TGF-β1 and ICAM-1 significantly increased in HG group. The expression of TGF-β1 and ICAM-1 was suppressed dose-dependently in moderate and high concentration adiponectin group(10~20μg/ml). Conclusion: The high glucose could inhibit the proliferation activity of RMC and promote the expression of TGF-β1 and ICAM-1. The low concentration adiponectin (2.5μg/ml)could promote the proliferation activity of RMC ,while moderate and high concentration adiponectin(10.0,15.0,20.0μg/ml) could inhibit the expression of TGF-β1 and ICAM-1.The results suggest that adiponectin maybe protect the kidney of patients with DM by decreasing the expression of TGF-β1 and ICAM-1,and provide a new method to prevention and cure complications of diabetes.
