Objective:To observe effects lowing the rate of solution which contains fluorocarbon on liver's reperfusion injury during the process of preserving liver by cold machine perfusion and find the objective rate of solution which can preserve liver best by clod machine perfusion.Methods:Using isolated perfusion of rat liver model,Wister rat livers were randomly preserved by cold machine perfusion.The rate of solution with contains fluorocarbon were controlled at 0.4,0.2,0.1 ml/(min·g).The content of aspartate transaminase(AST) alanine transaminase(ALT) and endothelin(ET) in reperfusion solution were tested after liver preservation 18h.The liver's cell and sinusoidal endothelial cell(SEC) under light microscopy and election microscopy were also observed morphologically.At the same time,set up the no cold machine perfusion's rat liver as the normal test.Comparing the above parameters with normal test,we can know the effect of lowing rate of solution on the liver which preserved by cold machine perfusion.Results:AST,ALT and ET were both higher in the 0.4,0.2 and 0.1ml/(min·g) group than in the normal group.There were no remarkable differences between 0.2 and 0.4ml/(min·g) group(p>0.05).There were remarkable differences between 0.1 and 0.4ml/(min·g) group(p<0.05).The histopathologic changes of liver in 0.4 and 0.2 ml/(min·g) group were lightener than in 0.1 ml/(min·g) group.Conclusion:During the process of preserving liver by cold machine perfusion, the rate of 0.1 ml/(min·g) perfluorochemical oxygen carrier solution should increase the injury of liver's cell and SEC.At the same time,the reperfusion injury of liver is seriously.
