| Objectives:The purpose of this study is with randomized controlled methods,by observing the differents among the four groups of SD rats of clean grade: cytochrome C oxidase activity and mitochondrial permeability transition pore' s activity to explore the relations between the mitochondrial oxidative damage and osteoporosis.It also added in the osteoporosis research on muscle, as well as providing a foundation for traditional Chinese medicine that through the maintenance of muscle mitochondrial function and delay apoptosis,to prevent osteoporosis.Methods:Select 52 Clean 6-month-old female healthy non-pregnant SD rats. First,52 rats were randomly divided into blank group and the surgery group, respectively,16 and 36.After the 36 rats ovariectomized,respectively, randomly selected 6 rats from the each group.Detect the whole body bone mineral density to determine the success of Osteoporosis Model.The left 30 rats of the surgery group were randomly divided into 3 groups:the traditional Chinese medicine(TCM) group,the Nilestriol group,and the Model group Each group has 10 rats.The blank group and the Model group were given distilled water orally, The TCM group were given the traditional Chinese medicine which can bushenjianpi to fill the stomach according to 4.8g/kg(which is 10 times as much as clinical dosage).The medicine needs constant volume to the same density by the distilled water,namely 10ml/kg,and was given once a day. The Nilestriol group were given Nilestriol Img/kg to fill the stomach. Confected Nilestriol into suspension,(The density is 0.167mg/ml).It was given to the Nilestriol group once a week.The other time gave the Nilestriol group distilled water lOml/kg to fill the stomach.3 months later,executed the mouse.Take the musculus quadriceps fexoris and gastrocnemius,and placed in the -80℃refrigerator to preserve.After that extracted mitochondria, and detected the activity of skeletal muscle' s mitochondrial permeability transition pore and cytochrome C oxidase.Finally,use statistical software SPSS13.0 data to build up a database for statistical analysis of the four groups on the activity of cytochrome C and mitochondrial permeability transition pore and determine the differences.Results:Mitochondrial permeability transition pore shows that the blank group and the Nilestriol group had significant differences(P<0.05) with the Model group on the ratio of 30min/0minOD and 60min/0minOD;While the Chinese group had no statistically significant difference with the model group.The black group were significantly different(P<0.05) with the Model group on the activity of cytochrome C oxidase,but the TCM group and the Nilestriol group had no difference with the Model group.Conclusions:Ovariectomized rat' s MPTP activity was significantly enhanced Skeletal muscle mitochondrial oxidative damage increased.The Chinese Medicines has no significant effect to MPTP activity but Nilestriol may have effected the activity of MPTP Nilestriol may stop mitochondrial oxidative damage and apoptosis by contraling the activity of MPTP.The Chinese medicine and Nylestriol had no significant effect on the activity of cytochromeC.The Chinese medicine and Nylestriol may stop mitochondrial oxidative damage by contraling the activity of cytochromeC,but its effect is not significant. |
