Objective:To observe the NF-κB/p65,COX-2 expression after transiently transfecting human gastric carcinoma cell SGC-7901 using siRNA,to approach the effect of RNAi targeting NF-κB/p65 on the proliferation and apoptosis of gastric carcinoma cell.Methods:Three groups were divided,siRNA group,negative control group and blank group,accordingly transfect the group with specific NF-κB/p65 siRNA,NC siRNA,①12,24,48 hours after transfecting SGC-7901 cells with specific NF-κB/p65siRNA or NC. siRNA,MTT method was used to detect the proliferation on the cells.②24,48 hours after transfection,cell cycle and apoptosis were carried out on the cells via flow cytometry.③The level about NF-κB/p65 mRNA,COX-2 mRNA of the cells were detected by RT PCR.④Immunnohistochemistry was taken to detect the expression of the protein about NF-κB/p65 and COX-2 in three groups.Results:The group which has transfected the specific NF-κB/p65 siRNA was compared with the other two groups,after transfectecting the specific NF-κB/p65 siRNA,①the cells of NF-κB/p65siRNA group grew much more slowly,the cell proliferation of NF-κB/p65siRNA group was obviously suppressed(p<0.05),②the cells were mainly retarded in G1 cell cycle,the amout of cells stayed within G0-G1 phase were obviously increased,cells stayed within S and G2-M phases were obviously decreased,the apoptosis rate was increased(p<0.01),③The expression of NF-κB/p65 mRNA and COX-2 mRNA in the specific siRNA group was decreased by a large rate(p<0.01),but negative control group and blank group had no obvious differences(p>0.05).④The expression of NF-κB/p65,COX-2 protien were down-regulated highly,therefor,with no obvious differences between the negative control group and blank group. Conclusions:Silencing NF-κB/p65 can effectively inhibit the proliferation of the cells about human gastric carcinoma,the cells were retarded in G0-G1 phase and were induced to apoptosis,the effect of Silencing NF-κB/p65 is probablely via the way of down-regulating the expression of COX-2.
