Correlation Between 18F-FDG Uptake And The Expression Of Glut1,Ki-67 And P53 In Esophageal Cancer And Its Clinical Values

Objective:To investigate the relationship between 18F-fluorodeoxyglucose (18F-FDG) uptake(L/B) and expression of glucose transporter-1(Glut1),Ki-67 and p53 in patients with esophageal cancer, and to analysis the relation between L/B and staging, infiltration depth, lymph node metastasis and prognosis.Methods:35 patients with esophageal cancer were evaluated with 18F-FDG SPECT examination before opertion. Relative value was calculated with esophageal carcinoma and normal esophageal tissue.The expression of Glut1, Ki-67 and p53 in the tumor tissues and 10 normal esophageal tissue was detected by sterpavidin peroxidase(SP) immunohistochemistry.Results:There were abnormal 18F-FDG uptaking regions on SPECT imaging of esophagus in 35 patients, which were confirmed histologically as the primary esophageal squamous cell carcinomas. It was 82.9%(29/35)that L/B was higher than 2.0(4.86±2.08), and it was 17.1%(6/35) that L/B was lower than or equal to 2.0(1.65±0.18. All the 35 tumors tested were Glut1 positive(80.0±19.1)% of tumor cell membrane, All the 10 normal esophageal tissues tested were Glut1 negative. Ki-67 antigen expression was all exceed 10%, p53 expression was 85.7%(30/35). Ki-67 expression was less than 10% and p53 expression was negative in all 10 normal esophageal tissues. There were significant differences in the expressions of Glut1 and Ki-67 among the esophageal cancer and normol esophagus( P<0.05). Correlations were found between Glut1, Ki-67 expression and 18F-FDG uptake(r=0.786,P<0.05;r=0.732,P<0.05). Correlations were not found between p53 expression and 18F-FDG uptake(r=0.261,P=0.130). Correlations were found among L/B and Staging (P=0.047), infiltration depth(P=0.003), There was no correlation between L/B and lymph node metabasis(P=0.624), it appeare to be correlated with L/B and survival rate (P<0.05).Conclusion: There were widespread expression of Glutl,Ki-67 and p53 in esophageal cancer tissues. There were significant differences in the expressions of Glut1,Ki-67 and p53 between the esophageal cancer and normol esophagus(P < 0.05) . Correlations were found among Glut1,Ki-67 and p53 and development and invasion metastasis. Relations were found between Glut1,Ki-67 expression and 18F-FDG uptake, but there was not a relation between the expression of p53 and L/B, and the L/B may be used to indirectly evaluate the energy metabolic level and proliferative capacity of esophageal cancer. L/B showed much values in M stage and prognosis judgement, and it's value was limtited in T and N stage in patients with esophageal cancer.
