The development of the quality control of TCM(traditional chinese medicine) based on the modern analytical methods,and the establishment of fingerprint of TCM quality control technology,which is in line with the characteristics of TCM,have been the key to ensure that TCM has become a safe,effective and controllable,and to achieve the realization of modernization of TCM.In this paper,to study Ophiopogon japonicus(Thunb.)Ker-Gawl and Saposhnikovia divaricata(Turcz)Schischk.we used HPLC,off-line 2D-RPLC/RPLC and FTIR in the fingerprint of TCM quality control studies.First of all,five binary solvent gradient reversed-phase conditions have been designed to analyse the n-butanol fraction of Ophiopogon japonicus(Thunb.)Ker-Gawl.Using computer simulation with CSA software systemto acquire a better separation condition.Then using HPLC to analyse the sample with this condition.The actual experimental data and simulation data are analysed and compared,find the peak time and peak number is tally to 92%.On this basis,13 samples of Ophiopogon japonicus(Thunb.)Ker-Gawl from different origins are used to creat a fingerprint spectra.In the second,based on the construction ofβ-CD and Waters XBrigde C18 column 2D-RPLC/RPLC analysis system,the n-butanol extract of Ophiopogon japonicus(Thunb.) Ker-Gawl is studied.The separation resolutionis significantly increased,and the total number of the peak also increased significantly.It will be more convincing to apply this method to the research and quality control of chemical constituents of TCM.Then,GC-MS was used to analysis the volatile oils of Saposhnikovia divaricata (Turcz)Schischks which were collected from four different places.The results show that,the chemical consititutes and their relative percentage contents of the volatile oills of Saposhnikovia divaricata(Turcz)Schischks in defferent areas vary greatly.At last,we used FTIR to explore Ophiopogon japonicus(Thunb.)Ker-Gawl and Saposhnikovia divaricata(Turcz)Schischk.The experimental results show that:the chemical compositions contained in the samples from different origins are different.So the absorption wave number of the functional groups and absorption intensity are varied.In this paper,optimization of computer simulation of conditions with HPLC and 2D-RPLC/RPLC are combined to study the quality control of the n-butanol extracts of Ophiopogon japonicus(Thunb.)Ker-Gawl.Using GC-MS to analyse the volatile oil of Saposhnikovia divaricata(Turcz)Schischks,and using FTIR to analyse Ophiopogon japonicus(Thunb.)Ker-Gawl and Saposhnikovia divaricata(Turcz)Schischk.The experimental results show that,the results of the analysis method built in this paper are satisfactory,the application should be promoted.
