Objective:To study the expression and meaning of P2X1,P2X3 receptors in rat bladders with OAB-symptom which were introduced by different causes,and to investigate the possible relationship of P2X1,P2X3 receptors with OAB-symptomMethods:To make rat's models which have OAB-symptom produced frome different causes as experimental group,normal rats as control group.Experimental group divides into five sub-groups:First,5 rats with OAB-symptom produced fi'ome bladder outlet obstruction after 3 weeks.Second,6 rats with OAB-symptom produced frome bladder outlet obstruction after 6 weeks.Third,5 rats with OAB-symptom produced frome bacterial cystitis.Fourth,6 rats with OAB-symptom produced frome hight-level-Spinalized.Fifth,6 rats with OAB-symptom produced frome cyclophosphamide(CYP)-induced cystitis.Cystometrogram(CMG) was performed in experimental group to get rats with OAB-symptom.Getted every groups' bladders to make pathologic chips and make HE,Masson stain.At the same time,We performed immunohistochemistry to localize of P2X1 and P2X3 receptors.We obtained OD of these immunohistochemical pictures by Image-pro-plus6.0,then analyzed them by analysis of variance.Results:Maked rat's models 5 groups:First,10 rat's models with bladder outlet obstruction for 3 weeks which produced OAB-symptom rats 5(50%).Second,10 rat's models with bladder outlet obstruction for 6 weeks which produced OAB-symptom rats 6(60%).Third,6 rat's models with bacterial cystitis which died land produced OAB-symptom rats 5(100%).Fourth,6 rat's models with hight-level-Spinalized which produced OAB-symptom rats 6(100%).Fifth,6 rat's models with cyclophosphamide (CYP)-induced cystitis which produced OAB-symptom rats 6(100%).In Morphology, bladders with outlet obstruction after 3,6 weeks and hight-level-Spinalized show smooth muscle cells hypertrophy,fibrous tissue increased and filled in the interspace between the smooth muscle layer.Bladders with bacterial cystitis and cyclophosphamide(CYP)-induced cystitis Show changes of bladder inflammation.The resaults of immunothistochemistry shows P2X1,P2X3 express in bladders of control group and experimental group.The quantity of express is much higher in experimental group than control group(p<0.05).The expression of P2X1 receptors were predominant in bladder detrusor and P2X3 receptors were predominant expressed in bladder urothelium.At same time,the condition of expression among some groups is different.Conclusions:The OAB-symptom preduced frome different factor have different pathologic changes.The expressive quantity of P2X1,P2X3 in the OAB-symptom bladders is much higher than normal rats.At the same time,P2X1 receptors expression were predominant in bladder detrusor and P2X3 receptors were predominant expressed in bladder urothelium.The changes of the expressive quantity of P2X1,P2X3 in bladder perhaps have a great role in the cause of OAB-symptom.The changes may be the common molecular biologic basic in OAB-symptom produced from different factors.The deeply research of the molecular biologic effection of P2X1,P2X3 in the reason for OAB-symptom perhaps offers a new theoretical basic for the manufacture of clinic medicine which can release the symptom of OAB-symptom.
