Objective:The Investigation of the effect of aerobic exercise on cell apoptosis and expressions of Bcl-2 and Bax protein in myocardium and skeletal muscle from aging model rats,so as to explore the application value and mechanisms of aerobic exercise on anti-aging From the point of apoptosis.Methods:To establish aging rat model,Subjects of present study were female SD rats of 21-month and 3-month in age,3-month-old rats were randomly divided into young group(n=8) and model group(n=24),The rats model group were treated with daily celiac injection of saline and D-gal(500mg/kg.d) respectively for 42 days. Randomly divided established aging model rats into control group and exercise groups which include 6 times/wk exercise group and 3 times/wk exercise group,exercise groups were trained by swimming for 12 weeks,90min for each time,all animals were killed 48h after the last training session.Apoptosis of myocardium and skeletal muscle was detected using TUNEL staining.The expressions of Bcl-2 and Bax in myocardium and skeletal muscle were quantified by western blotting,and to account the Bcl-2 / Bax ratio.Results: 1.TUNEL staining showed that compared with youth group the natural aging group and simple model group(randomly selected 8 from the model group) showed markedly increased brown nuclear myocardial apoptotic cells.2.Compared with control group,the apoptosic index of myocardium and skeletal muscle in exercise groups was obviously decreased(P<0. 01),the expression of Bcl-2 increased(P<0.01) and expression of Bax decreased(P<0.05).The ratio of Bcl-2/ Bax increased(P<0.01). Compared with 6 times/wk group,the apoptosic index of cardiomyocyte in 3 times/wk group was lower(P<0.05),expression of Bcl-2 increased (P<0.05),expression of Bax decreased(P<0.05),and the ratio of Bax/Bcl-2 was increased(P<0.05).Conclusion:1.By intraperitoneal injection of D-galactose for 42 days,The cardiac myocyte apoptosis index of 3-month-old SD rats is close to that of 22-month-old SD rat,Show that the model successfully.SD rat model of aging can be a substitute for the natural aging SD rats for the follow-up pilot study.2.Long-time aerobic exercise can increase Bcl-2/Bax ratio.It is beneficial to inhibiting apoptosis of cardiomyocyte,to advancing cardiomyocyte survive and to retarding its aging,to prevent and reduce the incidence of cardiovascular disease in the elderly. 3.Long-time aerobic exercise can increase Bcl-2/Bax ratio.It is beneficial to inhibiting apoptosis of cardiomyocyte,Can effectively reduce the trend of skeletal muscle atrophy result from Loss of muscle fiber.4.Through the Comparison of h 6 times / week,3 times / week (alternate days) the effect of every-other-day aerobic exercise was better than that of everyday aerobic exercise.
