Purpose: To demonstrate immunohistochemical expression of MUC4.MUC5AC MUC16 and NF -κB/p65 proteins in ovarian epithelial cancer, epithelial borderline tumors ,epithelial benign tumors and normal ovarian tissues,to establish their correlation with clinical stages,histological types,grades and lymphoid metastasis of ovarian cancer..Methods: 93 paraffin specimen of 52 ovarian epithelial cancers,11 ovarian epithelial borderline tumours,20 ovarian epithelial benign tumors and 10 normal ovarian tissues were collected. IHC were used to detect the protein of MUC4,MUC5AC,MUC16 and NF-kb/p65,using the anti- MUC4,MUC5AC,MUC16 and NF-κB/p65 monoclonal antibodies.Results and Conclusion: (1)There were different expression rates of MUC4, MUC5AC, MUC516 and NF-κB/p65 in ovarian epithelial tumor. The positive expression rate of MUC4 in normanl,benign, borderline and malignant ovarian epithelial tumor were 0,10%,36% and 80%; MUC5AC were 0,50%,36% and 13%; MUC16 were 0,20%,54% and 61%; NF-κB/p65 were 10%,20%,73% and 92%.(2)The positive expression of MUC4, MUC5AC and NF-κB/p65 in ovarian cancers was significantly higher than that in normal ovarian tissue, epithelial benign tumors and epithelial borderline tumors (p<0.01).It suggests that the three molecules may relate to the oncogenesis of ovarian cancer.(3)The positive and over expression of MUC4, MUC516 and NF-κB/p65 in moderately,poorly differentiated tissues was significantly higher than that in well-differentiated tissues(p<0.05).It suggests that moderately,poorly differentiated cancer cells proliferate actively.(4)The positive and over expression of MUC4, MUC516 and NF-κB/p65 in lymphoid metastasis of ovarian cancers was significantly higher than that in unlymphoid metastasis ones(p<0.05).It suggests that the three molecules may have certain connection in the progression of lymphoid metastasis,the detecting of the three molecules may have great value in predicting lymphoid metastasis and prognosis of ovarian cancers.(5)The positive expression of MUC4, MUC16 and NF-κB/p65 inⅢ-Ⅳstages of ovarian cancers was unsignificantly higher thanⅠ-Ⅱstages of ovarian cancers(p>0.05). (6) The positive expression of MUC4 and NF-κB/p65 in CA125-higher group of ovarian cancers was unsignificantly higher than that in CA125-normal group of ovarian cancers(p>0.05). The positive expression of MUC16 in CA125-higher group of ovarian cancers was significantly higher than that in CA125- normal group of ovarian cancers(p<0.05). (7) We compared the expression profile between MUC4 and MUC16,notably MUC4 expresion was significantly higher than MUC16(p<0.05),which together with other indexes,could be a potential candidate marker for early detection of epithelial ovarian carcinoma. (8) There was a positive correlation between MUC4 and NF-κB/p65(r=0.71, p<0.01),and a positive correlation between MUC16 and NF-κB /p65(r=0.530,p<0.01). MUC5AC and NF-κB/p65 are no correlation.(r=-0.058, p=0.578). Mucins expression may be under the regulation of NF-κB/p65.
