Objective To investigate the Levels and Effect of Vascular endothelial growth factor(VEGF),tumor necrosis factor-α(TNF-α) and InterLeukin -6(IL-6) after balloon injury in rat carotid artery.Methods 60male Sprague-Dawley rats were divided into the control group (A,no injury),the model group(B,the simple injury),thalidomide small doses group(C,the simple injury,100 mg·kg-1·d-1 of thalidomide was administered after conventional carotid artery denudation injury) and the high doses group(D,the simple injury,200 mg·kg-1·d-1 of thalidomide was administered after conventional carotid artey after arten injury).The rats were killed at 7,14,28days.Serum concentrations of VEGF were measured by enzyme-linked immunosorbent assay(ELISA) and serum concentrations of TNF-α,IL-6 were measured radioimmunoassay immediately.Results Serum concentrations of VEGF,TNF-αand IL-6 were significantly higher in B,C and D group throughout the experiment in comparison with A group(P<0.05);Compared with the B group,serum concentrations of TNF-α,IL-6 and VEGF were markedly reducedin A,C and D group in different times after injury(P<0.05);Compared with the C group,Serum concentrations of VEGF,TNF-αand IL-6 were also markedly reduced in D group in different times after injury(P<0.05).Conclusions TNF-α,IL-6 and VEGF were high expressed obviously after arterial injury.We found that thalidomide,effectively inhibits Serum IL-6,TNF-αand VEGF in balloon-injured rat carotid arteries.The level of three factors were correlated with the dose of thalidomide.
