Part oneThe expression of HIF-1a,nNOS after acute cerebral hypoxia-reoxygen in rats and drug interference studyObjective:To observe the expression of HIF-1a,nNOS in cerebral hypoxia rats and the effect by ginsenoside Rd interference as well as to explore its possible mechanism.Methods:60 adult wistar rats were divided into 3 groups:acute hypoxia group, hypoxia pretreatment group and ginsenoside Rd interference group;to observe the change of hippocampus cell shape and the expression of HIF-1a,nNOS in every group.Results:At immediate reoxygen time,we can found a few expression of HIF-1a,nNOS protein which mostly consisted in hippocampus cell,much of them in cytoplasm;as time processed,the level of HIF-1a,nNOS gathered the peak at reoxygen 4h;the level of HIF-1a markedly decreased at reoxygen 9h;and the level of nNOS markedly decreased at reoxygen 24h.There is statistical differences for AOD values between control groups and interference groups(P=0.009<0.05).There is no statistical differences between two interference groups(P>0.05).Conclusion:Acute hypoxia can cause the expression of HIF-1a,nNOS in rat hippocampus cells and its expression has time dependence;at the same time,hypoxia pretreatment and ginsenoside Rd interference can make the expression of HIF-1a,nNOS decreased;it suggested that these pretreatment measures may have protective action for acute hypoxia cerebral injury,the regulation of Ca2+influx and the inhibition of oxygen free radicals may be one of its mechanism. Part twoThe expression rule of PARKIN after acute cerebral hypoxia-reoxygen in ratsObjective:To observe the expression of PARKIN protein in cerebral hypoxia rats.Methods:adult wistar rats were divided into 3 groups:acute hypoxia group,hypoxia pretreatment group and ginsenoside Rd interference group;to observe the change of hippocampus cell shape and the expression of PARKIN protein in every group.Results:PARKIN protein expresses in normal oxygen status,chiefly in the cytoplasm of hippocampus cells,its expression decreases in hypoxia status and has time dependence.The results of hypoxia pretreatment and ginsenoside Rd showed that the expression of PARKIN is decreased less than that of the noninterference group,the AOD values of different time are statistically different(P=0.009<0.05);there is no statistical differences between two interference groups(P>0.05).Conclusion:Acute hypoxia can cause the expression of PARKIN protein decrease in rat hippocampus cells and its expression has time dependence,the hypoxia pretreatment and ginsenoside Rd interference can make it express more than that of the noninterference groups,this may produce protection to acute hypoxia cerebral injury,the mechanism may be related with the dysfunction of UPS under hypoxic conditions.
