Objective Explore the expression of HIF-2α(hypoxia-inducible factor-2α), TAMs(tumor-associated macrophages),VEGF-C(vascular endothelial growth factor-C) in breast carcinoma.Aimed at understanding the relationship among them and their roles in lymphatic metastasis of breast carcinoma,which can provide a reference for prognosis in breast carcinoma.Methods The expressions of HIF-2α,TAMs and VEGF-C were detected by immunohistochemistry in 53 human Breast Carcinoma samples and 20 normal breast tissue samples.Results(1) The expression rate of HIF-2αwere 34%(18/53) in breast carcinoma and 5%(1/20) in normal breast tissue.There was statistical difference between them(x2=9.015,P<0.05).The positive expression rate of HIF-2αwere 71.9%(22/31) in lymph node metastasis group,18.2%(4/22) in no lymph node metastasis group.There was significant difference between them(x2=14.347, P<0.05).(2) The expression rate of CD68 were 60%(32/53) in breast carcinoma and 15%(3/20) in normal breast tissue.There was statistical difference between them(x2= 11.98,P<0.05).The positive expression rate of HIF-2αwere 71.4%(23/31) in lymph node metastasis group,38%(8/22) in no lymph node metastasis group.There was significant difference between them(x2=7.59,P<0.05).(3) The expression rate of VEGF-C were 58%(31/53) in breast carcinoma and 10%(2/20) in normal breast tissue.There was statistical difference between them(x2=13.78,P<0.05).The positive expression rate of HIF-2αwere 70.97%(22/31) in lymph node metastasis group,27.27%(6/22) in no lymph node metastasis group. There was significant difference between them(x2=9.86,P<0.05).(4) The expression rate of HIF-2αin breast carcinoma had no correlation compared with patient age,tumor size,menopause status,(P>0.05 respectively),but was significantly related to tumor histological stage and axillary status.(P<0.05).(5) A significant difference was found in terms of expression of HIF-2αbetween the samples with TAMs either positive or negative(P<0.01),The same difference was found in VEGF-C(P<0.01).(6) Not only the HIF-2αexpression but also the expression of VEGF-C in breast carcinoma was related to the expression of TAMs.Conclusions(1) HIF-2αpromote lymphatic vessel formation by upregulating the expression of VEGF-C in breast carcinoma.Detection of HIF-2αcan be used as monitoring indicators in breast carcinoma prognosis(2) HIF-2αand TAMs have synergistic role in lymphatic metastasis of breast carcinoma(3) The VEGF-C participates the course of neonatal lymphatic vessels.TAMs infiltrating breast carcinoma can promote lymphatic metastasis of breast carcinoma by secreting VEGF-C.TAMs in breast carcinoma is one of the important factors in lymphatic metastasis.
