Objective:Investigate the therapeutic effects of the XINSHEN 1 HAO on gastric mucosal injury induced by chronic stress in the rat,and it's therapeutic mechanisms.This is good to excavate ancient prescription,and to provide a new idea and some experimental evidence in order to apply it in clinic better.Methods:1.This experiment used 60 male SD rats.They were divided into 6 groups by random(10 per group):the normal group,the model group,the western medicine group(ranitidine group),the small dosage group of XINSHEN 1 HAO,the middle dosage of XINSHEN 1 HAO,the large dosage of XINSHEN 1 HAO.2.In addition to the normal group from the rest of the groups keeping the first two weeks of stress begin to stimulate,at the beginning of every morning,the folder tail 30min-1 hour,drilling barrels 3 hours,fixed limbs 2 hours,water 24 hours, cut fresh 24 hours,one stimulating way per day,alternating 15 days.We administered the 1.5mg/ml Ranitidine Capsules fluid to the Western medicine group,2ml/100g body weight.(about 0.15g,0.3g,0.6g/100g crude drug) the XINSHEN 1 HAO fluid to the XINSHEN 1 HAO group,2ml/100g body weight distilled water to the model group and the fake operation group for about 15 days.3.In the remedial process,we observed the rat's appearance,spirit,behavior.15 days later,we executed all the rats.Then we anatomized the rats,and separated the stomach.We select the pituitaries,to check their histopathological change.Using method of immunohistochemical assay to detect express of c-fos,caspase-3 of gastric mucosa.Using method of RT-PCR to detect express of c-myc of gastric mucosa from six randomly selected rats per group.4.We analyzed the data by analysis of variance and q-test.Results:1.During the experiment,every group mouse outrage,bray,tug,worry,frighten and showed a gradual reduction in appetite,slow gain in weight,yellow color,and so on.That basically corresponds macro behaviors of disharmonious function of qi activities. The therapeutic groups are better than the model group in appetite,weight,hair color and quality. 2.It is succeed in adopting the method of combination of many kinds of chronic stress to produce the mouse model of gastric mucosal injury,stomach-tissue of different group of morphology changed differently.Gastric wall of model group of degree of injury is most severity.To compare traditional Chinese medicine group with model group,there is discrepancy statistical significance for UI(P<0.01).3.Compared with normal group,c-fos positive expression was significantly higher in model group(P<0.01).c-fos positive expression in XINSHEN 1 HAO -treat groups and western medicine group was significantly lower than that in model group(P<0.01)4.Compared with normal group,caspase-3 positive expression of was significantly higher in model group(P<0.01),caspase-3 positive expression in XINSHEN 1 HAO -treat groups and western medicine group was significantly lower than that in model group(P<0.01)5.There are no difference between the normal group and the model group.But compared with normal group,c-myc expression has advanced tendency in the model group,c-myc expression in the small and the middle dosage group of XINSHEN 1 HAO was significantly more than those in model group(P<0.01).Conclusions:1.It is succeed in adopting the method of combination of many kinds of chronic stress to produce the mouse model of syndrome(Qiji disorder ) and disease(gastric mucosal injury),The traditional Chinese medicine compound Xinshenyihao have a good effection to this model.It reflected differentiation of TCM.2.The expresstions of c-fos,caspase-3in the model have a distinctness change,it clews that they participate in the gastric mucosal injury and disharmonious function of qi activities.3.XINSHEN 1 HAO has many effects on gastric mucosal injury induced by chronic stress in the rat.XINSHEN 1 HAO can cut down the expresstions of c-fos,caspase-3 and enhance the expresstion of c-myc to promote the repair of gastric mucosal injure.It provides theoretical basis for Chinese medicine treatment of gastric mucosal injury by Qi disorder.
