Objective To study the biomechanics characteristics,interval porosity and adhesion performance of bovine acellular bone matrix and investigate the feasibility as a natural scaffold of bone tissue engineering.Methods The acellular bone extracellular matrix was prepared by method of 10% NaCl joint 1%TritonX-100.To observe the mean pore diameter and interval porosity of the ABECM by decalcifying section and HE stain.Giving a mechanical load test to the specimens used the mechanical equipment ElectroForce3200.The test index include:elastic strength destroy load and elastic modulus.By the Cytometry,the adhesive rate of the ABECM was tested using the osteoblast inducted from the marrow mesenchymal stem cells(MSCs) were co-cultured with ABECM in vitro.Results The method of 10%NaCl joint 1%TritonX-100 could completely remove the cell components of the ECM.Compared with the control group,the experimental group ECM had no significant damage after Acellular.The porosity of ABECM was 70.15±2.98%.The mechanical testing showed that no significant effect on mechanical properties of the ECM by the method of 10%NaCl joint TritonX-100.The ABECM had satisfactory adhesion performance with osteoblast cultured in vitro,the adhesion rate was 62.38%.Conclusions The bovine acellular bone matrix is an ideal natural scaffold material for bone tissue engineering.ABECM has satisfactory mechanical properties and the closest to the physical structure in diameter and porosity.Adhesion properties meet the requirements of bone tissue engineering scaffold.As a natural scaffold,ABECM has broad research and application prospects.
