Back ground:There is mutual checks-balances relation ship between tumor cells and immune system.A comprehensive exposition of the molecular mechanism of the two is one of the more cutting-edge in the life sciences currently.The immune system try to control an eliminate any mutant cells through series of mechanisms.During the same time the tumor cells also have a series of protection mechanisms to suppress or offset the function of the immune system in order to form tumors.Comprehensive and in-depth understanding of the interaction between the two systems,analysising the structure and function of the involved protein will provide a new target for cancer biotherapy. In early stage of breast cancer,there may have a local lymph node metastasis,which can damage local tissue and become the birthplace of the transfer of other organs.According the current view,breast cancer is a systemic disease,metastasis and recurrence is a major impact factor for long-term survival.There is few opportunities for local recurrence after radical surger.So distant metastasis is a major obstacle to long-term survival,with lymph node metastasis as the main means.Therefor there is close relationship between lymph node metastasis and the prognosi of breast cancer patiens.Discussion the structure and function of associated protein for lymph node metastasis of breast cancer will have signifance imprtant for suppressing distant metastasis,determeting prognosis and choicing treatment.Research purpose:Find out the cytokines related to lymph node metastasis of breast cancer.Research method:Collected 160 cases venous blood of patholocially confirem breast cancer patients before chemotherapy.Use Luminex liquid protein chip technology to detect the quantity of 29 kinds of cytokines molecules in serum.The 29 kinds of cytokines are IL-1β,IL-2,IL-1ra,IL-4,IL-5,EGF,IL-6,IL-7,TGF-α,Fractalkine,IL-8,IL-10, IL-12p70,IL-13,IL-15,IL-17,IL-1α,INF-γ,G-CSF,GM-CSF,TNF-α,Eotaxin, MCP-1,sCD40L,IL-12p40,MIP-1α,MIP-1β,IP-10,VEGF.Use chi square test to analysis the different of positive radiot between between lymph node negative and positive patients;use independent sample T-test method to analysis the different of the quantity between the two group.For positive patients,divide into the following three groups 1-3,4-9,≥10,according their positive lymph node number.So analysis the relationship between the expression intensity of lymph node metastasis-associated cytokines and the number of positive lymph node.Further more,divided the patients into two groups accoding whether or not distant metastasis.Analysis the relationship between selected cytokines and distant metastasis.In order to verify the repeatability of the experiment,detected IP-10 and MCP-1 in a new 160 serum samples,which we selected 80 cases from the above 160 cases randomly and 80 new cases.The results:The positive radiot of IL-1ra and MIP-1βof node-negative patients is higher than that of node-positive patients(P<0.05).The level of IP-10 and MCP-1 which is significant elevated in node-positive patients,is also have a inverse correlation with the numble of positive nodes(P<0.05).The level of MCP-1,Eotaxin and Scd40L in metastased patient is lower than non-metastased patients(P<0.05).The result from the new 160 serum sanpples is also has significant difference(P<0.05) between lymph node-negative and positive groups,with repeatability of the experiment.Conclusion:Serum level of MCP-1,IP-10,IL-1ra,MIP-1βhave a close relationship with lymph node state,and the level of MCP-1,Eotaxin,sCD40L is lower in metastased patients than non-metastased.Our results support an extend analysis of serum cytokine profiles for the potential development of predictive panels in diagnosis,staging and monitoring cancer treat.
