Objective To explore the expression and possible function of Bmi-1 and Mel-18 in gastric adenocarcinoma cells in vitro and in vivo,We set out to explore the expression and clinical significance of Bmi-1 and Mel-18 gene in gastric adenocarcinoma..Methods Real time RT-PCR was used to detect the expression of Bmi-1 and Mel-18 in cancer tissue and corresponding para-cancer normal tissue in 71 cases of gastric adenocarcinoma and in gastric adenocarcinoma cells,The association between Bmi-1 and Mel-18 expression and the clinicopathological parameters of the tumors was analyzed.The association between Bmi-1 and Mel-18 expression was also analyzed.Results The analysis revealed significantly increased expression of Bmi-1 in 5 of 9 carcinoma cells in comparison with normal cell and decreased expression of Mel-18 in 4 of 9,increased expression of Bmi-1 in carcinoma tissue in comparison with para-cancer normal tissue in 24 (33.80%) of 71 gastric adenocarcinoma cases and decreased expression of Mel-18 in 36(50.70%) of 71.The increase of Bmi-1 expression was significantly correlated with,tumor size,lymph node metastasis and T stage(P<0.05).The decrease of Mel-18 expression was significantly negatively correlated with lymph node metastasis and clinical stage(P<0.05).our study showed that the expression of Mel-18 is negatively correlated the expression of Bmi-1(P<0.05).Conclusion Our results demonstrated that Bmi-1 and Mel-18 may play crucial role in tumorigenesis and progression of gastric adenocarcinoma. It is possibly to list Bmi-1 and Mel-18 as biomarkers for predicting the prognosis of gastric adenocarcinoma.
