ObjectiveTo investigate the influence of Fatty infiltration in Pancreas on the function of the Islet Isolation and Purification in rat.MethodsTo explore the establishment of the differential degree diet-induced obesity model in the weaning male SD rats with high fat diet and normal diet.60 male rats are divided into 4 group which are normal group,mild group,moderate obesity and sever obesity group(n=15),with normal diet after adaptively feeding for two weeks.To observe the degree of fatty infiltration by pancreatic tissue slices obtained from the tail of pancreas in kinds of groups from kinds of groups.The remain pancreatic tissue digested with CollagenaseⅤand purified with ficoll400 are used to detect the function and activity of Islet,and further assess the influence on the function of the Islet isolation and purification by the degree of Fatty infiltration in pancreas.Results1.The differential degree diet-induced obesity model is established successfully with high fat and normal diet.The body weight,Lee's index were measured after feeding 14 weeks,compared with the contrary group,the difference among these data has statistical significance(P<0.01),according to the difference of degree of Fatty infiltration in pancreas by observing pancreatic tissue slices of differential groups.2.The difference between mild obesity,moderate obesity and sever obesity and normal groups,has statistical significance(P<0.01).Compared with the contrary group, the mean islet equivalents and function of purified Islet from sever obesity group are decreased obviously.Compared with mild obesity groups,the mean islet equivalents purified islet from moderate obesity group increased obviously after digestion and purification.Viability and potency assays in vitro are uncovered no difference among kinds of groups.Conclusions1.The differential degree diet-induced obesity model can be successfully established with the admixture of high fat and normal diet,moreover,the differential degree fatty infiltration in pancreas also can be implemented accordingly.2.Compared with normal rats,the rats of mild obesity and moderate obesity are suitable for islet isolation and transplantation,particularly,moderate obesity rats are more suitable.Their use could increase the size of the islet donor pool.But the sever obesity are not suitable for islet isolation and transplantations.
