Effect On Insulin Resistance Rat Model Of Polycystic Ovarian Syndrome Treated With Puerarin

Objective:To study the change of estradiol(E2),progestone(P), Testosterone(T),Luteinizing hormone(LH),follicle-stimulating hormone(FSH),Insulin(INS) in the serum and the Glucose level of blood in polycystic ovarian(PCO) rat models.To establish polycystic ovarian rat model and investigate the relationship between PCOS and insulin resistance.To observe effects of puerarin on glucose,serum levels of insulin,estradiol,progestogen,T FSH LH and HOMA index before after therapy With Puerarin.and initially approach it' s mechanism of action.Female SD rats with regular estrous cycle aged 8-9 weeks were divided into control group(n=18),PCO model group(n=25) after feeding 4 days.All rats were made model besides control group.The control group were given common animal feeds and common water; The rats in the model groups were induced by Poretsky,s method for PCO modeling and maintained for 6 weeks on high-fat-sugar diet.Both groups' bodyweight,ovary size and weight,HE staining,glucose tolerance,plasma INS,E2,T,P,FSH,LH levels.The ovary were taken down,the moiety were maken patho-section and another moiety were reserved. After confirming the model being suitable for thePCOS research,The modeling were divided into model group(n=8),treatment group(Puerarin)(n=9) and control group(n=8) randomly,followed by administration of Sodium Chloride 1ml/100g/d or Puerarin 100mg/kg/d.After 28 d treatment,the rats were sacrificed and the level of INS,E2,T,P,FSH,LH.in the serum were detected by Elivision TM Plus.The blood glucose and oral glucose tolerance test(OGTT) were detected simultaneously.Then HOMA index was calculate.The ovary were taken down,and the moiety were maken patho-section and another moiety were reserved.Results:Macroscopic observation found that the control groups' ovarial surface seems ruddy and some corpus luteum,the model making groups' ovarial surface were pale and the volume increasing,and small amounts corpus luteum,Observing by microscope that the ovarian follicle extend and oocyte or corona radiata lose,and the granular cell layers decreased to 2ˉ3 layers and artially follicular atresia,and the control group 8～9 layers.These indicated the model making is successful(Fig).Compared with normal control group,The ovarian colour were red and the corpus luteum can be found of the treatment group,there are many corpus luteum and all ovum be found by microscope.The Saccular dilatation decreased obviously or abolition,granular cell layer increased,follicular fluid and ovarium mound can be found in dominant follicle(Fig).compared with other groups,the endometrium of the Puerarin group had no significant changes in shape,but the numbers of glandular organs decrease in PCOS group.In control group,glandular organ of the endometrium presented cluster-form arrangement glandular cavity is big and have much pleat,glandular epithelium appeared some secretory vacuoles.In model group,the glandular organ were scattering. glandular cavity is tiny and straight.Compared to control group the puerarin treatment group had no distinction greatly.The model making groups rats' body weights was significantly higher than the nomal control group(P＜0.05),the puerarin group rats' body weights was significantly decreased than the model groups(P＜0.05).these indicated magnolol could impove the PCO rats functional condition.Compared with normal group,the padding was wetter obviously in the cage of model group,and the levels of plasma INS,T,LH,Glucose increased,and significant glucose intolerance abnormality,so as the HOMA index.But FSH was no statistics defference during the groups,the results indicated the model of insulin resistance was successful.In the puerarin treatment group,the INS,T and LH level declined(P＜0.05),which were lower than the levels of model group,and OGTT,HOMA were also improve apparently.the results indicated that the puerarin could improved rats' serum hormone levels and insulin resistance.Conclusions:Therapeutic action is significant of puerarin to the modeling rats of PCO,That it can alter ovarian volume,colour,and pathology state,significantly decreased obesity rats' body weights of PCO and improve their functional condition;puerarin could decreased rats' serum hormone levels(INS,T,LH) and retrieved insulin resistance symptome. Therapeutic action of Puerarin to the modeling rats of PCO possible by raising insulin sensitiveness and cut down INS,blood glucose,IR.Because the insulin resistance is intercommunication and interrestrict with other hormone,So serum T,LH also degrade.
