Objective Bronchial asthma is a bronchial disease which is characterized of a chronic inflammatory process in the airway of asthmatic individuals,and which is lead by the immunologic response.Airway inflammation in asthma is associated with the local activation of T lymphocytes in the airway mucosa.In allergic asthma these activated T lymphocytes are predominantly of the Th2 cytokine phenotype.T lymphocytes play an important role in the development of chronic airway inflammation with bronchial asthma.By observing the cytokines of type of Th1(IFN-γ,IL-12) and type of Th2(IL-4) which in lymph,bronchoalveolar lavage fluid(BALF) and blood in asthmatic rat,to clarify the dynamic changes of proliferation and differentiation of T lymphocytes during the lymph in the course of airway inflammation,as well as the relationship among lymph,BALF and blood.This paper aims to explore the immunologic pathogenesis of asthma,and the effect of lymph in the immunological regulation.We are going to find a new way to prevent and cure asthma.Methods Asthmatic rats were established in the study.Rats were sensitized and challenged by ovalbumin(OVA).Lymph,BALF and blood were taken and IL-4, IFN-γand IL-12 were assessed at 0hour before challenged,2,6,10,24,48hours after challenged,with enzyme-linked immunosorbent assay method.We are going to research the relationship of cytokines among three aspects at different time points.Results 1.We successfully establish the lymph draining experimental model of asthmatic rat,which witnessed by clinical,pathology,and differential count of BALF.2.The levels of IL-4,IFN-γ,and IL-12 in lymph,BALF,and blood at different time points of control group.1) The average concentrations of IL-4 in lymph,BALF,and blood were 13.21±1.52,8.44±1.22,5.72±1.16(pg/ml).IFN-γwere 94.30±10.41,74.21±8.73, 44.17±4.69(pg/ml).IL-12 were 22.81±2.15,15.77±1.26,8.04±1.06(pg/ml).There were no significant differences in three cytokines at different time points(P>0.05)2) The levels of IL-4,IFN-γand IL-12 in lymph were significantly higher than in BALF and blood at different time points of 0,2,6,10,24,48hours(P<0.01).The levels of IL-4,IFN-γand IL-12 in BALF were significantly higher than in blood at different time points of 0,2,6,10,24,48hours(P<0.05).The levels of IL-4, IFN-γand IL-12 in lymph were 1.6,1.3,1.4 times of BALF,and 2.3,2.1,2.8 times of blood.3.The comparisons of IL-4,IFN-γ,and IL-12 in lymph,BALF,and blood at different time points of asthmatic rats.1) The average concentrations of IL-4 in lymph,BALF,and blood were 31.99±6.31,22.42±2.45,17.42±4.36(pg/ml).IFN-γwere 57.80±12.93,42.57±9.92, 19.08±6.17(pg/ml).IL-12 were 10.53±4.01,4.61±0.97,3.14±0.84(pg/ml).2) The levels of IL-4,IFN-γ,and IL-12 in lymph were significantly higher at different time points of 2,6,10,24hours than in 0hour(P<0.05).The level of IL-4 in lymph increased 2h after OVA challenge,and reached its maximal level at 10h.IL-4 in BALF and blood increased 2h,and reached its maximal level at 24h. IFN-γ,IL-12 in lymph increased at 2h,and reached its maximal level at 6h.IFN-γ, IL-12 in BALF and blood increased at 2h,and reached its maximal level at 10h.3) The level of IL-4 in lymph was significantly higher than in BALF and blood at different time points of 0,2,6,10,24hours(P<0.05).The levels of IFN-γand IL-12 in lymph were significantly higher than in BALF and blood at different time points of 0,2,6,and 10hours(P<0.05).The levels of IL-4,IFN-γand IL-12 in lymph were 1.4,1.4,2.3 times of BALF,and 1.8,3.0,3.3 times of blood.The levels of IL-4,IFN-γand IL-12 in BALF were 1.3,2.2,1.5 times of blood.4) The levels of IL-4 in lymph,BALF and blood in asthmatic rats were significantly higher than in control group at different time points of 0,2,6,10,24, 48hours(P<0.01).They were 2.4,2.7,3.0 times of control group.The levels of IFN-γand IL-12 in lymph,BALF and blood in asthmatic rat were significantly lower than in control group at different time points of 0,2,6,10,24,48hours(P<0.01).IFN-γwere 1.6,1.7,2.3 times of control,IL-12 were 2.2,3.4,2.6 times of control group.Conclusions1.We have successfully established the lymph draining experimental model of asthmatic rat,which witnessed by clinical,pathology,and differential count of BALF. At the same time,we improve the method of lymph collection on the basis of others.2.We discuss the disposition features of IL-4,IFN-γ,and IL-12 in lymph,BALF, and blood at 0,2,6,10,24,48hours.The results showed the levels of IL-4,IFN-γ, IL-12 in lymph,BALF,blood become lower and lower one by one.They indicate lymph is the most localized district of cytokines,and it can reflect the immune response and inflammation degree of body.3.The comparisons of IL-4,IFN-γ,and IL-12 in lymph,BALF,and blood at 0,2, 6,10,24,48hours of asthmatic rats.The results showed effect of Th1/Th2 cytokine was significant in lymph.The level of IL-4 in lymph of asthmatic rats was significantly higher than in control group,and the levels of IFN-γand IL-12 were significantly lower than in control group.The three cytokines emerged earlier in lymph than in BALF and blood.The phase variation was significant.Our research show cytokines disorder maybe come from lymph system at first.We suppose cytokines in lymph maybe could reflex the inflammation degree of asthma much earlier and more exact.
