| Rubus chingii is plant of Rosaceae, distributed in Zhejiang, Jiangsu, Anhui, Jiangxi, Fujian, Guangxi. The unripe fruits of it have been used as a food and tonic in traditional Chinede medicine. It has been used in China to treat impotence and seminal emission as a tonic for thousands of years.BackgroundRubus chingii has been used in China to treat impotence and seminal emission as a tonic for thousands of years. But modern study especially in chenmical conetituents and pharmacological mechanism is not enough. So, the phytochemical study and the pharmacolgogical active about curing kidney-yang deficient have a large significance on developing of rubus chingii.ObjiectiveIt is the arm of this work to find the bioactivity substance of Rubus chingii through chemical constituent and pharmacodynamics research.MethodIn the basis of literature investigation, we collect the fuits of Rubus chingii from Zhejiang province, and identify the botanic origains. We choose hydrocortisone to make the model of kidney-yang deficiency and took weight of the mouse, concentration of testosterone, histopathological change of kidney and testicle as index to study whether the extract of Rubus chingii can alleviate the kidney-yang deficiency symptom.We Screened the active part of Rubus chingii through giving the kidney-young mice petroleum extract, dichlormethane extract, n-BuOH extract, water extract. And detect the weight of the mouse, concentration of testosterone, histopathological change of kidney and testicle to determin which part has the best active. We study the chemical constituent of the active part of Rubus chingii, and use the mean of organic solvent extraction, chromatograph of silica gel and recrystal isolate compound. The structures were charaxterized by spectral methods.Result and discussionThe mice injeced intraperitoneally with hydrocortisone acetate has the symptom of minimum weight, the concentration of testosterone decreased, the cells of kindey diminution and spermatocyte decreased in testicle. The extract show the effect on kidney-yang deficient mice inducesd by hydrocortisone.The petroleum extract, dichlormethane extract, n-BuOH extract and water extract can allevate the kidney-yang deficiency syndrome induced by hydrocortisone. The n-BuOH extract has the best activity. So, we confirm n-BuOH extract as the effective part of the extract .We isolated five compounds from the n-BUOH extracts, and the structure of four compounds was characterized by spectral methods. One new compound was 3,5,9-trihydroxy-7,8-dihydrocyclopenta[g]chromene-2,6-dione, named"Fu-Pen-Zi-Su"by author, and three known compounds were tiliroside, Kaempferol, cuercetion.We isolate eleven compounds from the dichlormethane extracts, and the structure of nine compounds was characterized by spectral methods. They are hexacosanol,β-sitosterol, 4-hydro-3-methlbenzal dehyde, 4-hydrobenzal dehyde, Oleanolic acide, Stigmast-5-en-3-ol,oleate, H-2-indenone,2,4,5,6,7,7a-hexahydro-3-(1-methylethyl)- 7a-methyl, 4-hydroxy-3-methoxybenzoic acide, liballinol. Exceptβ-sitosterol, Oleanolic acide, 4-hydroxy-3-methoxybenzoic acide, all the other compounds were obtained from Rubus chingii for the first time. |
