The Dynamical Expression Of VEGF And AQP4 In Cerebral Ischemia Rats's Hippocampi

Objective: To investigate the expression of Vascular endothelial growth factor (VEGF) and aquaporin4 (AQP4) at different times in cerebral ischemia rats's hippocampi,then explore the effects of VEGF and AQP4 on the edema come into being and disappear.Method: One hundreds and eight Healthy SD rats (W=200~250g) were randomly divided into 2 groups, the control group and the experiment group. And then the rats were subdivided into 18 groups(six rats for each)at an interval of 3h,6h,24h,2d,3d,1w,2 w,1m and 2m respectively. The experiment group were ligated double Carotid and the control group are only remover double carotid sheath . To utilize the immunohistochemical method to examine the protein expression of VEGF and AQP4 in different times of in ischemic rats's cerebral. The results were analyzed by means of imagic motic Med 3.0 Analyzer,the data was handled with SPSS 13.0 statistic software.Results: 1. The AQP-4 selectively expressed in the cell membrane, The VEGF expressed typically in endothelial cytoplasm, astrocytes .2. The expression of VEGF in The experiment group significantly increased after 6h and had its peak at 2d.Then it decreased afte 1 week,and return to normal after 2 weeks.3. The expression of AQP4 in The experiment group significantly increased after 6h and had its peak at 2d.And return to normal after 1 week.4. By observing the gray density,The expression of VEGF was significantly different at 6h,24h,2d,3d and 1week compared with the control group(p<0.05) ,The expression of AQP4 was significantly different at 6h,24h,2d,3d and compared with the control group(p<0.05) .5. Furthermore the VEGF expression was significantly positive correlated with the AQP4 expression (P<0.01,r=0.846)determined by spearman correlation.Conclusion:At the earlier period of hippocampi in the ischemic rats's cerebral, the expression of AQP4 and VEGF were strongly incresed.At the later period,the expression of AQP4 and VEGF were down-regulated.The Brain edema of has a result of expression alteration of many cell factors and the VEGF with the AQP4 may play a decisive role, and they are maybe interrelated...
