Aim:Airway inflammation is involved in asthma and COPD, as well as in fiber cystis degeneration (CF) and bronchopulmonary dysplasia (BPD). Chronic airway inflammations always accompany with the structural change of airway wall and parenchyma, affect respiratory function severely. And the airway structure, inflammatory cells and matrix protein change frequently, and result in more complex etiopathogenesis and clinical manifestation. Glucocorticoids is considered to be the most efficiency in treating asthma, can't deteriorate the pathogenesis and the immunity disproportion, and weakenes the protecting immunity and initiates general side effect. Chinese medicine is more effected, with more targets, weak side effect, being good at precluding and aiming directly at the etiopathogenisis, and is unsuccedaneous in the prevention and cure of asthma, and therefore, to study the effect and mechanism of Chinese medicine therapy is not only conduced to elucidate the connotation traditional Chinese medicine in asthma, but also significative in clinical therapy .Methods: Experiment 1 Pharmacological experimental study of Kechuanan capsule on antitussive and expectorant effects50 KM mice were divided into 5 groups randomly. The normal group was given NS intragastric administration, the positive control group was given Benproperine (54mg·kg-1), 3 experiment groups (low group, middle group and high group) were given Kechuanan capsule correspondingly (406mg/kg, 812mg/kg and 1624mg/kg). Every group was given the drug once everyday for 10 days. And put mice in groups in YLS-8A multifunction instrument for inducing tussis and conducting wheeze once for 5s, 10s and 2min on day 11, day 12 and day 13 correspondingly after given medicine. And every mouse'scough times in 2 min were record. Experiment 2 The inhibition of Kechuanan capsule in asthma mice and its mechanism60 BALB/c mice were divided into 6 groups randomly: the normal group, the asthma group, the positive control group, the Kechuanan low group, the Kechuanan mid group, the Kechuanan high group. Every group was allergized on day 0, 7, 14 for 3 times except the normal group. From day 15, every mouse in all groups was put in a 20 cm×20 cm×15 cm closed container for 30 min inhaling 3% OVA to stimulate for 14 days, except the normal group for NS. Every group was given the drug (406mg/kg, 812mg/kg and 1624mg/kg) correspondingly for 14 days. The positive control group was given dexamethasone (2.0 mg/kg) by intraperitoneal injection. The normal group and the asthma group were given NS. All animals were killed in 24 hours after the last suscitation. To select the lung, and then put the right lung in 10% formaldehyde for immobilization, and the left lung were made into tissue homogenate to detect the concentration of MDA and NO, and detect the concentration of IgE in blood serum and the concentration of IL-17 and EOS in BALF.Experiment 3 Therapeutical effect of Kechuanan capsule on chronic bronchitis model mice and its possible mechanism60 KM mice were divided into 6 groups randomly: the normal group, the asthma group, the positive control group, the Kechuanan low group, the Kechuanan mid group, the Kechuanan high group. To establish the model by cigarette and ammonia water to induce the chronic tracheitis. Every group was given the drug (406mg/kg, 812mg/kg and 1624mg/kg) correspondingly for 15 days. The positive control group was given aminophylline (69.9 mg/kg). The normal group and the asthma group were given NS. All animals were killed in 24 hours after day 15. To select the lung, and then put the right lung in 10% formaldehyde for immobilization, and the left lung were made into tissue homogenate to detect the concentration of MDA, NO, and MPO.Result:Experiment 1 All Kechuanan capsule groups can increase the trachea secretion of Phenolsulfonphthalein of mice. The Kechuanan capsule mid group effect is better than the NH4Cl group. The Kechuanan capsule can significant prolong the cough incubation period of mice, and reduce the cougf times significantly.Experiment 2 The content of MDA of the model group was more than that of the normal group. But there was no significant difference between the Dexamethasone group and the model group. All Kechuanan group could reduce the content of MDA accompany with the model group. And the content of NO of model group is higher than that of normal group. All content of NO of treatment group is lower than the normal group, but there no significant difference in the treatment groups and normal group. The IgE in model group serum was higher than that of normal group significantly, and all treatment group was lower than the model group. There was some dose-effect relationship in the Kechuanan groups. The IL-17 and EOS in model group BALF were higher than that of normal group significantly, and all treatment group was lower than the model group. There was some dose-effect relationship in the Kechuanan groups too.Experiment 3 The content of MDA of the model group was more than that of the normal group. But there was no significant difference between the Aminophylline group and the Kechuanan groups. All Kechuanan groups could reduce the content of MDA accompany with the model group. And the content of NO of model group is higher than that of normal group. All content of NO in treatment groups is lower than the normal group, but there no significant difference in the treatment groups and normal group. The MPO in model group was higher than that of normal group significantly, and all treatment group was lower than the model group. There was some dose-effect relationship in the Kechuanan groups.Conclusion:Kechuanan capsule can reduce the cough times induced by ammonia water, shows its preventing cough effect, and reduce the trachea phenolsulfonphthalein volume ejection, shows its effect of increasing the respiratory tract mucus secretion. The Kechuanan capsule can reduce the MDA content of asthma and chronic bronchitis, increase the content of NO, and reduce the MPO activity of chronic bronchitis shows that it can control the inflammation in asthma and chronic bronchitis. At the same time, it can reduce the IgE content in serum and IL-17 in BALF, this shows the effect of IgE and IL-17 in the process of asthma, and this prove that the anti-asthma and anti-inflammation effect of Kechuanan capsule is by regulating the balance of IgE and IL-17.
