| PartI Establishment of Rat Colitis Model Induced by Trinitro-benzene- sulfonic Acid/Ethanol and Immunocomplex.Background: Inflammatory bowel disease includes ulcerative colitis and Crohn's disease, which is the non-specific and inflammatory disease resulting from hereditary factor, environmental field and immunity agent in intestinal tract. Being unknown of etiology and pathogenesy, IBD is classified as one of refractoriness diseases by World Health Organization in modern time. Animal model of IBD has significanced in disease investigation of etiology, pathogenesy, development regularity, determination diagnositic and therapeutic tool and exploitation and research in new drug.Aims: To make colitis model induced by trinitro-benzene-sulfonic acid(TNBS) /ethanol and immunocomplex in rat,to expect as more ideal animal model of IBD and assess its value in research study of inflammatory bowel disease.Methods: 168 Sprague-Dawley rats, female and male hold a half each other, which were divided into four groups. (1) Fourty two rats as control group, isotonic Na chloride was instilled into rats colon according to 100mg/kg. (2) 42 rats as TNBS/ethanol group, which was instilled TNBS dissolved in 50% ethanol according to 100mg/kg. (3) 42 rats as Immunocomplex group, which antigen-emolsifier contained antigen of 8mg were injected into digiti pedis and fold inguen, interval time was two weeks. (4) 42 rats as TNBS/ethanol and Immunocomplex group, which were injected antigen-emolsifier firstly. We observed macroscopical, histological change, myeloperoxidase (MPO) activity and fecal occult blood test (OBT) at the 1st, 2nd, 3nd, 4th, 6th, 9th and 12th weekend after model established.Results: The mean of macroscopical, histological change, MPO activity, OBT of TNBS/ethanol and Immunocomplex rats were 5.8571±0.8431, 4.8571±0.3542, 3.8588±0.8625, 4.1429±0.6010 respectively, which was significently increased than control groups.Conclusions: TNBS/ethanol and immunocomplex induced rat distal colitis is an ideal model of IBD and may serve as a tool for investigation of the pathogenesis and pharmaceutical effect of IBD. Up to now, there is no an ideal animal model of IBD similar to IBD which happened in human in nosogenesis ,development regularity and pharmaceutical treatment etc. For the past few years, some studies from domestic and overseas have verificated that CD4+ T Cells plays key role in pathogenesy of IBD. Another studies have confirmed that abnormality of cytoimmunity and humoral immunity concurrence in IBD. Above which we mentioned are challenging to battle for animal model of IBD.Aims: From point of view of cytoimmunity and humoral immunity, the study was to confirm colitis model induced by TNBS/ethanol and immunocomplex compared with TNBS/ethanol which caused widespread attention in scholars all the better consistents with B lymphocyte and T lymphocyte intermingle infiltration in IBD. Further to verificate the colitis model expected as more ideal animal model of IBD and to assess its value in research study of inflammatory bowel disease.Methods: 168 Sprague-Dawley rats, female and male hold a half each other, which were divided into four groups. We observed change of CD4+ T Cells in peripheral blood, expression of CD4+ T Cells in colonic organism and content of IgG in blood serum at the 1st, 2nd, 3nd, 4th, 6th, 9th,12 th weekend after model established. Results: The mean percentage of CD4+ T Cells in peripheral blood, expression of CD4+ T Cells in colonic organism and content of IgG in blood serum of TNBS/ethanol and Immunocomplex rats were 44.81±9.1830% , 179.86±35.9546, 0.54±0.1205 respectively, which was significently increased than control groups.Conclusions: Further to verificate TNBS/ethanol and immunocomplex induced rat distal colitis is an ideal model of IBD and may serve as a tool for investigation of the pathogenesis and pharmaceutical effect of IBD. |
