Objective The aim of the study was to investigate the expression of TLR4 mRNA in primary chorionic trophocyte cell with or without the stimulation of LPS, the effect of TLR4 on the cell apoptosis and the secretion of TNF-αinduced by LPS.Methods Trophoblasts were isolated from chorionic membrane and cultured in vitro. Cultured chorionic trophocyte cells were treated with LPS for different time(0h, 2h, 12h,24h). The expression of TLR4 was detected by reverse transcription polymerase chain reaction (RT-PCR), the apoptosis of trophocyte cell was measured by Flow cytometry (FCM) and the secretion of TNF-αwas measured by enzyme linked immunosorbent assay (ELISA).Results TLR4 mRNA was expressed in Primary chorionic trophoblast cells and the expression of TLR4 mRNA increased when cells treated whith LPS. LPS could induce chorionic trophocyte apoptosis, and LPS-induced apoptosis could be blocked by pretreating with TLR4Ab. LPS could increse the secretion of TNF-α, and LPS-induced secretion of TNF-αcould also be blocked by TLR4Ab.Conclusion ultured chorionic trophocytes express TLR4 mRNA. TLR4 may play an important role in LPS-induced apoptosis and secretion of TNF-α.
