Objective: The osteosarcoma is one of the most common malignant tumors among young people.It is easy to palindromia and metastasize. The fatality rate and mutilation rate are exceeding high. It is severely detrimental to people's health. The occurrence and development of osteosarcoma is closely concerned with multiple oncogenes as well as anti-oncogenes. Apoptosis closely affect the occurrence of osteosarcoma, and The anti- apoptosis gene Bcl-2 and Bag-1 may are most important action in the occurrence,development, anticipation and prophylaxis of osteosarcoma. Most studies only detect the relationship of the expression of the single indicator Bcl-2 with the development of osteosarcoma ,but local authority has not been reported about the relationship of bag-1 in osteosarcoma and the interreaction relationship of Bcl-2 and Bag-1 in osteosarcoma . The research was to detect the expression of Bcl-2 and Bag-1 in osteosarcoma tissues using immunohistochemical method (SP), and to analyze the separate and associated expression of Bcl-2 and Bag-1 in osteosarcoma. Research the expression and the interreaction relationship to find a safety molecular indicatrix for patients to demonstrate the clinical Pathology characteristic,to provid theory evidence for developing the research of gene therapy of osteosarcoma.Methods: We randomly collected 30 osteosarcoma samples which were from amputated extremity, resected by surgery and biopsy in He Bei Medical University from 2001 to 2007. These cases were not received chemotherapy or radiotherapy . According to the diagnostic code of osteosarcoma ,they were confirmed by three pathology specialist after surgery.In these 30 patients, 19 cases were males, 11 cases were females.The ages range from 12 to 46 years old, and the average age was 22.4 years old. In these 30 samples, 13 samples were osteoblast , 9 samples were chondroblast, the others 8 samples were fibroblast.We made 15 normal bone tissues for control group in the First Hospital of Qin Huangdao, these tissues were not nontumorous or inflammatory and resected by surgery because of the treating requisition. .In these 15 patients, 9 cases were males, 6 cases were females, the ages range from 18 to 65 years old,the average age was 33.2 years old.In our study, we observed the expression of Bcl-2 and Bag-1 in osteosarcoma by immunohistochemistry method(SP) and analysed their relationship in osteosarcoma.The experimental data were annlyzed with Chi-square test and Spearman correlation by software SPSS12.0 and the standard of statistic significance was established asα=0.05.Results :1 Expression of Bcl-2 in osteosarcomaNormal bone tissues had low expression of Bcl-2, 3 in 15 normal bone tissues showed positive expression. Compared to normal tissues,19 in 30 osteosarcoma did show positive expression of Bcl-2,the expression of Bcl-2 in osteosarcoma tissues was much higher than that in normal bone tissues. There was significant difference between the two groups (χ2=5.3804,P <0.05). There are 8 osteosarcoma tissues in 13 patients with osteoblast had Bcl-2 expression. 6 osteosarcoma tissues in 9 patients with chondroblast had the Bcl-2 expression.5 osteosarcoma tissues in 8 patients with fibroblast had the positive expression . There were significant difference between the three groups (χ2 = 0.0635,P =0.9688 >0.05)。No correlation was observed with age(p=1.0000) or gender(p=0.6956).2 Expression of Bag-1 in osteosarcomOsteosarcom tissues had high expression of Bag-1,and had no expression or week expression of Bag-1.The results demonstrated that the expression of Bag-1 in osteosarcom (76.67%) was much higher than that in normal bone tissues (6.67%),the difference was very significant (χ2 =19.6875,P <0.05 ).The expression of Bag-1 in osteosarcom had no significant correlation with the type of osteosarcom tissues, the expression of Bag-1 in osteoblast tissues (76.92%) was as high as which in chondroblast (66.67%) and fibroblast (87.50%).The difference was not significant (χ2 = 1.0284,P >0.05). No correlation was observed with ageor gender. (p>0.05).3 The correlation between Bcl-2 and Bag-1 expression in osteosarcom. 23 cases had the positive expression of Bag-1 , 15 of them had positive expression of Bcl-2. 13 cases had the negative expression of Bcl-2,5 of them had negative expression of Bag-1. A positive correlation was observed between Bcl-2 and Bag-1 in osteosarcom(r=0.833,P<0.0001).Conclusions:1 The osteosarcom tissue had the higher expression of Bcl-2 , but had the lower expression of Bcl-2 in normal bone tissue. The expression of Bcl-2 had no association with the type of osteosarcom ,age and gender. The high expression of Bc2 has relation with the development, progression of osteosarcom.2 The osteosarcom tissue had the high expression of Bag-1 , but in normal bone tissue Bag-1 was weak expression or no expression . The expression of Bag-1 in normal bone tissues was much lower than that in osteosarcoma tissues. The expression of Bag-1 had no association with the type of osteosarcom ,age and gender.The high expression of Bag-1 has significant effect in the development,progression of osteosarcom.3 osteosarcom apoptosis is mediated by more than one genes .Compare to Bcl-2,the apoptosis regulator activity of Bag-1 is more dominant.4 The expression of Bcl-2 in osteosarcom had the higher expression of Bag-1. There was a significant result in statistics. This result suggested that the anti- apoptosis gene Bcl-2 and Bag-1 build up each other in the development of colorectal carcinoma.
