Background: In our protopased animal experiment, we had obsreved changes of mircocirculation after high-voltage electric burn by WX-9B poly-position microcirculation instrument,and demonstrated the existence of microcirculation disorder in burned animal modles.It is an important phenominon that leukocyte adhere to vascular endothelium after burn.It was mediated by intercellular adhesion molcule-1 (ICAM-1), lymphocyte founction-associated molecule-1(LFA-1) and macrophage surface antigen-1(Mac-1.Some experimental study manifested that morbidity and mortality of burn has mostly associated with leukocyte adhesion,however,correlated study is a relatively infrequent in electrical injury,so we design the high-voltage electrical burned experiment.Objective: To research the dynamic changes of ICAM-1,CD11b/CD18 and the mechanisms of microcirculation disorder,We may be offer theoretical foundation for the clinical therapy,decrease mortality of patients after high-voltage electrical injury. Methods:1. Group:144 healthy grown up S-D rats are randomly divided into three groups,high-voltage electrical burned experiment group(experiment goup),high-voltage electrical burn UTI reatment group(treatment group)and fake high-voltage electrical burn group (control group),48 rats of each group.regardless of sex,The female rabbits have not been pregnant. the time phase of these groups are divided into six different phases,ahead(T0),immediately(15 min)(T1),1h(T2),2h (T3),4h(T4),8h(T5),8 rats of each group.2. Preparetion:to weigh the rats,number them ,moult the rats in inner sides of the left upper limbs,right lower limbs and belly with 8% sulfureted natrium liquor,then use normal sodium to wash them and dry naturally,put them in 24℃-26℃laboratory,no food and free water for a night.3. Duplication of high-voltage electric burn model: Anaesthetic rats of electrical burn experiment group and UTI treatment group are burned by the 10,000 volts high-voltage electric system for 5 seconds,they are duplicated full-thickness electrical burn with body tissues that touch with the electrodes, we immediately inject UTI to rats of treatment group through abdomen, normal sodium to experiment group.The progress of control group is the same as the electric burn group,but there isn't electric current in the body of control group rats.4. Collection and detection:In different time phases open pectoral cavity take suction blood about 5ml from rats'heart and fractionate 4ml and 1ml,the former centrifuge with 3000r/min about 10 minutes,suction supernatant,and then preserve in ultra low temperature freezer;we extract neutrophil from the latter,then prepare cell suspension and immobilization.4.1. The serum content of sICAM-1of the rats were measure by enzyme-linked immunosorbent assay(ELISA) at different time phases during early electrical burn.4.2. Flow cytometric analysis(FCM) was used to investigate mean fluorescence intensity(MFI) of CD11b/CD18 at different time phases during early electrical burn, MFI reflects the expression levels of CD11b/CD18.5. Analysis of Statistics: statistically analysis all the data with SPSS16.0.all the dates should be expressed with x±s.we apply one-factor analysis of variance(ANOVA) among groups.It has significant statistically meaning if p<0.05.Conclusion:1. the changes of sICAM-1 in the serum(1) The serum levels of sICAM-1 haven't obviously change within the control group.(p>0.05)(2) The serum levels of sICAM-1 of experiment group increase obviously,especially T1 ,T2 in early postburn stage. compared with the control group, they have a significant increase.(p<0.05)(3) The serum levels of sICAM-1 of UTI treatment group increase obviously also in early postburn stage. They have a significantly higher than the control group. (p<0.05) (4) The study reveals a significantly decrease of serum levels sICAM-1 of UTI treatment group than the experiment group.(p<0.05)2. The changes of CD11b/CD18 expression levels(1) The expression levels of CD11b/CD18 haven't obviously change within the control group.(p>0.05)(2) The expression levels of CD11b/CD18 of experiment group increase obviously in early postburn stage,Compared with the control group, they have a significant increase.(p<0.05)(3) The expression levels of CD11b/CD18 of UTI treatment group increase obviously also in early postburn stage. They have a significantly higher than the control group .(p<0.05)(4) The study reveals a significantly decrease of expression levels of CD11b/CD18 in UTI treatment group than the experiment group.(p<0.05)Conclusion:1 soluble Intercellular adhesion molecule-1(sICAM-1) and macrophage surface antigen(Mac-1) have increase after high-voltage electrical burned experiment group and UTI treatment group, play a important role in microcirculatory disorder.2 The study indicates sICAM-1 and CD11b/CD18 of UTI treatment group higher than experiment group, but lower than the control group still. sICAM-1 and CD11b/CD18 might be helpful in preservation and relief microcirculatory disoeder after high-voltage electrical burn.
