Objective:To observe the effect of soybeanisoflavone and grape seed proanthocyanidin on apoptosis in pancreatic isle cell of Type two diabetes ratsMethods:SPF SD rats were randomly divided into two groups:normal control,the group animals received normal animal feeds and disinfection water intragastric administration ,high fat diet group,were given hypso-lipoids feedstuff。After four weeks ,the blood was collected to detect the fasting blood glucose (FBG),the fasting insulin(FINS), triglyceride (TG) and total cholesterol (TC) to define hyperlipemia model.Then ,1% STZ was injected peritoneally into model.Those whose glucose level of tail vein blood were above 16.7mmol/L 72 hours after STZ injected were considered valid disabetes.And then the rats of high fat diet group were randomly divided into three groups : disabetes control group (B group), were given hypso-lipoids feedsuff and received disinfection water intragastric administration;SIF+GEPC group (C group, 250mg/kg.d), were given hypso-lipoids feedsuff and received SIF+GEPE 250mg/kg.d intragastric disinfection ; SIF+GEPE group (D group , 50mg/kg.d), were given hypso-lipoids feedsuff and received SIF+GEPC 50mg/kg.d intragastric disinfection .There are 6 rats in every group .The experiment last 4 weeks. all rats were killed, their pancreatic gland and blood serum were removed. Various parameters were measured as follows: each group rats glucose level of blood, at the end of experiment;the morphological study under light microscope;detecting the malondialdehyde (MDA),superoxidize dismutase (SOD), nitricoxide (NO)and reduced glutathione(GSH)level of serum:the apoptotic morphology under light microscope and Apoptosis ratio ;the protein expressions of Caspase-3 in pancreatic islet cell;the content of TNF-α, IFN-γin serum of rats.Results:(1) Acrossing to hypso-lipoids feedsuff the weight, FINS, TC,TG and insulin sensitivity index of high fat diet group rats were significintly increased ,compared with normal control (p<0.05). Those whose glucose level of tail vein blood were above 16.7mmol/L 72 hours after STZ injected were considered valid disabetes.(2)When diabetes is generate,the activity of SOD and GSH were inhibited, while the level of MDA and NO were promoted; but these abnormal state can be changed by SIF and GSPE.(p<0.05)(3) Acrossing to detect TNF-α,IFN-γ,Caspase-3 ,the apoptotic morphology and apoptosis ratio, we found disabetes control group rats islet cell apoptosis significintly increase(p<0.05). SIF and GSPE could protect pancreatic islet cell and decrease the level of above parameters (p<0.05).Conclution:(1) hypso-lipoids feedstuff and 1% STZ injected succeful set up type 2 diabetes model.(2) Disabetes damage antioxidation of body ,and lead to oxidative stress. Under oxidative stress irritating the body secreted apoptosis factor increase ,and then induce pancreatic islet cell apoptotic .(3)SIF+GSPE could anti-apoptosis through anti-oxygen and regulating the express of apoptosis factor.
