Objective To assess the protective effects of ulinastatin on acute lung injury induced by paraquat,investigate the influence of ulinastatin on NF-κB,TNF-αand IL-10 expression and evaluate the therapeutic effectiveness of ulinastatin for acute lung injury and its probably mechanism of action.Methods Experiments were performed on male Sprague-Dawley rats altogether 30,which were divided into 3 groups with 10 in every group:group NS(control group);group PQ(paraquat injury group);group UTI(group treatment with ulinastatin). Paraquat (80 mg/kg) was administered to the rats in group PQ and group UTI only once by oral. In the group NS,the same volume saline was administered.Rats in group UTI were injected into cavum abdominis of UTI 100 000u/kg at two time points(immediately and 12h after administer of PQ). In the group PQ,the same volume saline was injected.Rats in each group were killed by exsanguinations at 24h after administration of paraquat,and lungs were harvested for further examinations.Arterial blood gases were analyzed.Lung wet/dry weight ratio and protein in bronchoalveolar lavage fluid were measured.The pathomorphology changes of lung tissue were evaluated by H&E-stained lung sections.Immunohistochemistry was used to examine the expression of TNF-α,IL-10,NF-κBp50 and NF-κBp65.Expression of NF-κBp65mRNA was determined by reverse transcription polymerase chain reaction.Results Compared with rats in group NS,marked histological damage occurred in group PQ,characterized by diffused alveolar septum edema, Inflammatory cells infiltration,hemorrhaged in alveolar spaces and increased endothelial permeability.The W/D ratio of lung was significantly increased and the PaO2 was significantly lower in group PQ(Ρ<0.05).Rats in group UTI also had the above changes,but were milder in comparison with group PQ.Compared with rats in group NS, expression of TNF-α,IL-10,NF-κBp50,NF-κBp65 and NF-κBp65mRNA was significantly increased(Ρ<0.05),but in group UTI , expression of TNF-α,NF-κBp50,NF-κBp65 and NF-κBp65mRNA was significantly lower than those in group PQ(Ρ<0.05) and expression of IL-10 was increased(Ρ<0.05).Conclusion In paraquat-induced acute lung injury rat model,expressiong of NF-κB,TNF-αand IL-10 was increased. ulinastatin can attenuate the lung injury induced by paraquat and reduce the expression of TNF-αand NF-κB,increase the expression of IL-10.
