Object:To explore the expression of RECK and MMP-14 proteins in the human hepatic cancer with a series of clinical parameters, analyze its relation with development, metastasis and prognosis of liver cancer cases with its clinical significances, and find out their importance in haptic cancer. Methods:Immunohistochemical staining(IHC) was employed to determine the expression of RECK and MMP-14 proteins in 61 specimens of human hepatocellular carcinoma tissues and all their adjacent non-cancerous liver tissues and 18 normal liver tissues.Meanwhile, the relationship between expression of RECK and MMP-14 proteins in liver cancer tissues and various clinical parameters such as the age,the gender, the size of mass, the portal vein embolus, the metastasis of local lymph nodes, the degree of cirrhosis, the level of AFP or HBsAg and whether they had intact veins were analyzed.Cox proportional hazard regression model was employed to find the relationship between the two proteins and the prognosis sufferers. Results:The positive rate of RECK in hepatic cancer tissues was significantly lower than that in adjacent non-cancerous liver tissues[59.0%, 36/61 vs78.7%,48/61,P<0.05]and that in normal liver tissues[88.9%,16/18](P<0.05).As to MMP-14,the positive rate in liver cancer tissues was significantly higher than that in adjacent non-cancerous tissues[90.2%,55/61 vs 23.0%,14/55]and that in normal liver tissues(P<0.05).The positive rates of RECK and MMP-14 were significantly negatively correlated to each other. The expression of RECK protein was also significantly correlated with the clinical parameters such as the portal vein embolus and whether the tumors had intact veins(P<0.05),but were not significantly correlated with some other parameters such like the age,the gender, the size of the tumors, the metastasis of local lymph nodes, the degree of cirrhosis, nor correlated with the level of AFP or HBsAg.While the expression of MMP-14 protein was significantly correlated with the gender, the portal vein embolus, the level of AFP and whether the tumors had intact veins(P<0.05),but was not exactly connected with other clinical parameters(P>0.05).What is more,RECK was found to be much due to the prognosis of the cases and to be one of the independent protect factor of prognosis in liver cancers by the statistics methods of Cox model analysis. Conclusion:The expression of RECK protein may distribute to the infiltration of hepatic cancer, this effect may connect with its distinctive function with which RECK can restrain the expression of MMP-14 protein.
