Objective To study the apoptosis of Hela cells induced by stichopus japonicus sekenka (Sjamp)and its possible mechanism.Methods Hela cells were treated 72h by different concentrations of Sjamp,and were divided into four groups:0.8mg/ml group,1.2mg/ml group,1.6mg/ml group,0mg/ml group;The apoptosis of Hela cells was observed by electronic microscopy.The rate of apoptotic was determined by flow cytometry Annexin V-FITC/PI staining;The variation of intracellular Ca2+ concentration was detected by flow cytometry Fluo-3/AM staining;The change of mitochondria membrane potential was determined by flow cytometry rhodamine123 staining;Im-munohistochemical S-P method was used to detect the expression of Bcl-2 protein in the cells.Results Sjamp can induce the apoptosis of Hela cells obviously in dose-dependent manners;morphology changes of cell apoptosis such as karyopyknosis and conglomeration were observed by electronic microscopy;Flow cytometry showed the intracellur of Ca2+ concentration increased and the mitochondrial membrane potential of the cells decreased after the exposure to different concentrations of Sjamp;Immunohistochemical S-P method suggested that the expression of Bcl-2 protein in the cells decreased in concentration dependent manners.Conclision Sjamp could induce apoptosis of Hela cells,Ca2+ and Bcl-2 protein may play important roles in the regulation of cells apoptosis.The mitochondrial way might one of ways of Hela cells apoptosis induced by Sjamp.
