Objective:The experimental samples are human extraocular muscles.This study was designed to evaluate the express of nerve growth factor(NGF) and ciliary neurotrophic factor(CNTF) in the human extraocular muscles of congenital paralytic strabismus. Provide a theoretical foundation on the pathogenesis and treatment for congenital paralytic strabismusMethods:Collect 8 cases of strong-side and weak-side extraocular muscles from patients with congenital paralytic strabismus on 2008 January-October.And 4 pairs of the same normal muscles from patients with other ocular disease but normal extraocular muscles All these muscles were equally divided by sex,aged<3 years old.8 cases of strong-side and weak-side extraocular muscles from patients with congenital paralytic strabismus were used as the experimental group,4 pairs of the same normal muscles were used as the control group.All the muscles will be divided into three specimens.Taken by parts for semi-quantitative reverse transcription polymerase chain reaction(semi-quantitative reverse transcription polymerase chain reaction,RT-PCR), another part put into glutaraldehyde for electron microscope examination,the remainder put into formaldehyde for the conventional HE staining and immunohistochemical detection.Results:①Histological observation:Muscle fiber edema,disorders,vacuolar degeneration of mitochondria can be seen in the week-muscles.Fuzzy sarcomere structure,muscle fiber atrophy,the direction is irregular,or the whole muscle fiber showed vacuolization map,collagen fiber hyperplasia.Some of the strong-side muscle fiber sarcoma structure is still clear,but some of sarcoma disorder,and some lines disappear,mitochondria in muscle cell hyperplasia.Air bubble-like changes in sarcoplasmic reticulum expansion plans can be seen.Part of muscle fiber atrophy, interstitial widening,cell organelles increased.②Immunohistochemistry staining revealed that the staining of NGF and CNTF were lighter than corresponding normal muscles,both staining of strong-muscles were more obvious than normal.③RT-PCR results:NGFmRNA and CNTFmRNA of the weak-muscles were begun to descend than the normal.Both mRNA were up-regulated than the normal muscles.The difference was statistically significant(p<0.01).Conclusion:The expression of NGF and CNTF in the strong-muscles increased at both protein and mRNA levels.The expression of NGF and CNTF in the weak-muscles decreased at both protein and mRNA levels.It suggests that the express of NGF and CNTF have some correlation with congenital paralytic strabismus.Enhance or diminish NGF and CNTF gene expression may affect the role of ophthalmologic.It can provide a new theoretical foundation for the first research and treatment for congenital paralytic strabismus...
