Purposes:To investigate whether the bone marrow-derived mesenchymal stem cells can improve hyperglycemia and renal lesions in diabetic rats induced by low dose strptozotocin and high fat diet.Methods:1 Isolation and cultivation, identification of MSCs1.1 BMSCs were isolated from rat bone marrow from femurs and tibias under sterile condition.1.2 The surface markers: CD34, CD44, CD45,CD71, CD105 were analysed by flow cytometry.1.3 Vertification differentiation of MSCs into osteoblast by von kossa staining.2 Induction of type 2 diabetic rats.2.1 The diabetic model of Wistar rats was duplicated by injection of STZ intraperitoneally.2.2 The changes of blood sugar, insulin, triglyceride and were monitored real-time.2.3 The sections of pancreas were analyzed with immunofluorescent method.3 Transplantation of MSCs marked by BrdU3.1 Marking MSCs with BrdU.3.2 Injection MSCs into type 2 diabetic rats through tail vein.4 Measurement the effect of transplanted MSCs in the receptor.4.1 Monitoring the changes of blood sugar, insulin, triglyceride, serum adiponectin and urinary albumin.4.2 The sections of kidney and pancreas were stained by H-E and PAS.4.3 Analyzing the sections of kidney and pancreas with immunofluorescent method.Results:1 Cultivation, identification, labeling of MSCs: flow cytometry examination showed that MSCs, cultured in vitro, were expressed CD44, CD71 and CD105, but not CD34, CD45, and found anti-34, anti-CD45 negative and anti-CD44, anti-CD71, anti-CD105 positive.2 MSCs can be induced into adipoblast and osteoblast.3 The type 2 diabetic model were then affirmed when the blood glucose level more than 16.7mmol/L and stabilized for three days and the urinary albumin level more than 1.9mg/24h. 4 Possible existence of BrdU-labeled MSCs in Pancreas and kidney were assessed by Immunofluoresence. There is increasing of serum adiponectin(2.02±0.24μg/l;3.57±0.72μg/l), decreasing of insulin ( 206.08±3.18pmol/l ; 193.68±3.06pmol/l ) , blood glucose( 22.51±1.87mmol/l ; 17.02±1.27mmol/l ) , mean glomerular area(7.55±0.18μm2×103 ;5.21±0.45μm2×103), mean glomerular volume(8.12±0.029μm3×105;7.01±0.11μm3×105)and urinary albumin(1.95±0.21 mg/24h;1.30±0.15/24h). Therefor, transplantation of MSCs can result in beta - pancreatic islets regeneration and prevent renal damage in diabetic rats.Conclusions:The transplantion of bone marrow-derived stem cells could reside at the pancreatic site and renal of type 2 diabetic rats induced by STZ and high fat diets(induced diabetic rats), decrease insulin, blood glucose and excretion of urinary albumin , increase serum adiponectin ,in addition, it also can decrease insulin resistant and improve the glomerular hypertrophy, mesangial expansion, reduced the severity of glomerular sclerosis.
