Buyang Huanwu Decoction Re-development-Study On The Quality Control Methods Of Compound Capsule (Granula)

Objective Under the guidance of the traditional Chinese medicine theory, to ascertain the elementary active material of Buyang Huanwu decoction which is used to cure the Ischemic Stroke by following the " principal,assistant, adjuvant and guiding " formulation principle and compatibility rule;to extract the effective parts or efficacious constituents from each herb using the modern scientific technology,to mix them together as Chinese medicine Compound Capsule(granula),to establish the quality control methods, implement the expression of neotype complex prescription compound with digitization.Methods To determine the elementary active material of Buyang Huanwu decoction,abide by the " principal,assistant,adjuvant and guiding " formulation principle and compatibility rule of the traditional Chinese medicine formula,combining the correlation knowledge with science of Chinese medicine media and Chinese medicine of pharmacology.According to the different physico-chemical property of each active constituents,extract the herbs with different extraction methods.TLC was used to identify the effective part of Peach seed in the Compound Capsule(granula),TLCS was used to assay the contents of active monomer contained in effective part of Astragalus mongholicus;HPLC was used to assay the contents of active monomer contained in effective part of the Rootlet of Chinese Angelica,Red Peony Root, Chuanxiong and Safflower,and use the retention time for the qualitation,take astragaloside,ferulic acid,peoniflorin as quality control standard,use TLCS and HPLC to assay the contents of Compound Capsule(granula).Results In the Compound Capsule(granula),take the Astragalus saponin and polysaccharides as monarch,which have the effectiveness about invigorating qi for strengthening superficies;The effective parts of ferulic acid can enrich the blood and promote blood flow,in coordination with the monarch, so it is the minister;The effective parts of Peoniflorin,Gallic Acid, Ligustrazine and Glyceryl Oleate can enhance the rootlet of Chinese Angelica promoting blood flow and removing blood stasis,assisted by the Earthworm powder,enhance the effective part of Astragalus mongholicus invigorating the potential of vital energy and promoting blood circulation.The average extracta sicca rate of the effective part of Astragalus mongholicus is 35.47%, the Rootlet of Chinese Angelica is 45.05%,the Red Peony Root is 33.44%,the Chuanxiong is 28.67%,the Safflower is 34.83%,the average extracted oil rate of Peach seed is 42.30%.It can obtain 52.9458 gram Compound Capsule(granula) from a dose of prescription decoction.The TLC speckle of Peach seed is clear, the negative control is non-interference.The good linear correlation of astragaloside was observed from 1.015～8.120μg by TLCS,R~2 = 0.9993,and the average recovery of Compound Capsule(granula) was 97.91%,RSD = 2.07%;The good linear of ferulic acid correlation was observed from 0.055～0.275μg by HPLC,R~2 = 0.9998,and the average recovery of Compound Capsule(granula) was 96.73%,RSD = 1.42%;The good linear of peoniflorin correlation was observed from 0.497～7.952μg,R~2= 0.9997,and the average recovery of Compound Capsule(granula) was 98.92%,RSD = 1.77%.Conclusion The active material of Compound Capsule(granula) can interact and coordinate to bring about population effect of multi-strata,multi-target in human body,and then get the drug action,to achieve the destination of treating disease.The methods are simple,reliable,accurate,sensitive with good reproducibility and good specificity,They can be taken as the quality control methods of Compound Capsule(granula).
