Objective:To investigate the inhibitory effect of Dalteparin(a kind of low molecular weight heparin)on cell proliferation and expression of CyclinD1 mRNA of human ovarian cancer cell line SKOV3.Methods:The morphological change of SKOV3 treated by Dalteparin was observed under the microscope; the MTT method was employed to assay the growth inhibition rates at different concentrations of Dalteparin; the flow cytometer(FCM) was used to analyze the distribution of cell cycle; the semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) was used to assay the diversification of CyclinD1 mRNA expression.Results:When treated with the increasing concentration of Dalteparin and the prolonging treatment time,SKOV3 cells showed the following changes which were observed under the microscope:cell shrinkage,cell rounding,intercellular connections loose,some cells fell-off the wall,and even ruptured. When the concentration of Dalteparin was between 12.5IU/ml and 200IU/ml, the MTT assay at 24 hours and 48 hours showed a significant inhibitory action of Dalteparin, which can inhibit the proliferation of SKOV3 in dose-dependent and time-dependent manner in vitro. After the treatment of Dalteparin for 24 hours, the cell ratio of G0/G1 phase was significantly increased and S phase decreased (P<0.01), but the cell ratio of G2/M phase was not significantly changed (P>0.05). When treated for 24 hours,the expression of CyclinD1 mRNA was significantly reduced as the concentration of Dalteparin increasing (P>0.05).Conclusion:Dalteparin can inhibit the proliferation of ovarian cancer cell line SKOV3. The mechanism is probably that Dalteparin can arrest cells in G0/G1 phase and decreasing the expression of CyclinD1 mRNA.
