[Objective]To investigate the effect of the fibrin glue(FG) as a carrier compounded with BMP-2 on repairing bone defect and stimulating early period bone formation and to provide the experimental foundation for the clinical application.[Material and Methods]1.Preparetion of fibrin glue compounded with growth factor BMP-2 delivery system,and make the consistency of BMP-21ug/ml.2.Bone defects (8.00mm length,2.00mm width and 1.00mm depth)were created in 36 rabbits on the bilateral radius and were treated with three kinds of implantations.Experimental group:fibrin compounded with BMP-2;control group:fibrin;blank control group:no material.The animals were divided to 3 groups,then the defect were repaired with above three materials.More rabbits were taken to guarantee that 4 rabbits used to be examined in each group in each time point for the rabbits died and the failure model.3.Animals were respectively sacrificed at the end of 2 weeks,4 weeks and 8 weeks after operation.Then the speciments were made for gross observation,X-ray, histological section,immunohistoehemical method to detect the osteocalcin (OC),scanning electric microscope observation and energy dispersive analysis X-ray(EDAX) of element Calcium(Ca),Phosphonium(P) and Sulphur(S).SPSS 11.0 statistical package was used to deal with all the data.Data were analyzed by one -way ANOVA and paired t-test(a=0.05).There was significant difference when P<0,05 and highly significant difference when P<0.01.[Result]The bone forming activity and bone healing speed in experimental group were significantly better than those of the other groups by X-ray,histological section and scanning electric microscope observation.Immunohistochemical:The OC content were highest and the significant differences among the three groups in the second week(P<0.01).It was decreased that the expression amount of OC in local bone and bone callus organization in experimental group,and the most significant differences among the three groups appeared at the fourth week(P<0.01).There was no difference between the experimental group and the other groups at the eighth week(P>0.05).The result of EDAX showed that the content of element Ca and P of the newly formed bone increased obviously through the second and fourth week(P<0.01),while the contents of element S decreased obviously(P<0.01).After the eighth week,there was no obvious discrepancy of the amount of three elements between the newly formed bone of the experimental group and other groups(P>0.05).[Conclusions]It can be concluded that BMP-2 is a high-performance growth factor, and it can induce new bone and promote early bone formation.Fibrin glue(FG) has the good biology efficiency.It is a good release carrier compounded with BMP-2,and it could release growth factor gradually when it was analysed,so that BMP-2 can bring into full play of bone induction activity.The material with fibrin glue as a carrier compounded with BMP-2 could significantly accelerate the new bone forming,and it is beneficial for regeneration,mineralization and remodeling.It can shorten the recovery period,and stimulating early bone formation and osseointegration.
