Objective To evaluate the sensitivity of colchicine to the G422 glioma cell in vitro with colorimetric MTT assay.To inject different dosage of colchicine into mice with the G422 glioma and to observe whether this can prolong mice's life periods.Methods Experiment one:In vitro,sensitivities of 6 different concentrations of colchicine were determined by MTT method in G422 glioma which spread generation in the small mice body,which 6 kinds of concentration respectively are 0.03ug/mL,0.06ug/mL,0.12ug/mL,0.25ug/mL,0.5ug/mL,1ug/mL.The G422 glioma cells were plated in 96-well tissue culture plates and incubated for 48h with colchicine. Experiment two:To establish 80 mices subcutaneous glioma model,these will be inoculated with G422 glioma were randomly divided into 4 groups.According to colchicine 0.5,0.05,0.005 mg/kg·d,3 groups of 4 are divided into high,medium and low experimental group,and the other one group is the saline control group.20 mices in each group,sub-in after G422 inoculation 2d(equivalent to 48h),5d,will be injected 0.1ml of drugs at the original inoculation G422 subcutaneous,a total of 2 times.Results 1.The growth of the G422 glioma cell can be restrained by 6 different concentrations of colchicine,and its inhibition rate are separately 0.03ug/ml Group (36.41±4.16)%,0.06ug/ml Group(51.33±5.30)%,0.12ug/ml Group(65.47±2.77)%,0.25ug/ml Group(70.54±2.42)%,0.5ug/ml Group(75.38±1.10)%,1ug/ml Group(77.37±2.25)%.The rate of restraining growth of tumour cell increased with the concentrations of medicine.2.By colchicine treatment,tumor-bearing mices have extended and the average survival time were 44.28±1.07,43.79±1.64,43.09±1.39 days;the control group the average survival time was 28.28±1.77 days.More than the number of samples are compared by analysis of variance,P<0.01.Conclusions 1.The growth of the G422 glioma cell in vitro can be restrained by colchicine.In the 0.03-1ug/mL density scope,the growth suppression rate of the G422 glioma cell increases along with the medicine density advances.2.Chemotherapy of colchicine can significantly improve tumor-bearing with G422 mice's survival time and postpone tumor growth..3.The colchicine has the possibility to become one kind of new chemotherapy medicine to anti-glioma. |
