ObjectiveTo Compare the phenotype of dNK cells and the expression of IFN-γin dNK cells between the pregnant women with hypertensive disorder complicating pregnancy and the women with normal pregnancy. Investigating the relation between dNK cells and Hypertensive disorder complicating pregnancy. Methods(1) From August 2008 to March 2009, in the first affiliated hospital of JINAN University, twenty pregnant women with Hypertensive disorder complicating pregnancy were selected randomly as experimental group; at the same time, fifteen women with normal pregnancy were selected randomly as control group.(2) The Third-trimester decidual tissues were colleced through elective caesarean section.(3) Cell suspensions were prepared by an electromechanical dispersal and centrifugated using a standard gradient sedimentation technique. The phenotype of dNK cells were detected by flow cytometric analysis.(4) Exciting and cultivating the dNK cells with stimulant. Flow cytometry was used to detect the expression of interacelluar cytokine IFN-γin dNK cells.Results(1) In the control group, The mean of CD56brightCD16-CD3-dNK cells subgroup and CD56dimCD16+CD3-dNK cells subgroup were (59.5%±17.6%) and (8.9%±7.8%) respectively, the difference between the two subgroups was significant.(P=0.000<0.05) .(2) In the experimental group, The mean of CD56brightCDl6-CD3- dNK cells subgroup and CD56dimCD16+CD3-dNK cells subgroup were (55.0%±20.1%) and (9.4%±8.7%) respectively, the difference between the two subgroups was also significant. (P=0.000<0.05) .(3) Both in the control and experimental group, the cardinal phenotype of dNK cells were CD56brightCD16-CD3- (59.5%±17.6% vs 55.0%±20.1%), there was no statistical difference between the two groups: P=1.000>0.05.(4) In the control and experimental group, the mean of CD56dimCD16+CD3-dNK cells were (8.9%±7.8%) and (9.4%±8.7%) respectively, there was no statistical difference between the two groups: P=1.000>0.05.(5) There was no difference in the expression of CD 16 on dNK cells between the control and experimental group (13.6%±8.3% vs 12.9%±9.5%), P=0.56>0.05.(6) In the experimental group, the expression of intracellular cytokine IFN-γin dNK cells (32.6%±6.7%) decreased markedly as compared with those in the control group (20.4%±7.5%) (P=0.000<0.05).ConclusionsBoth in hypertensive disorder complicating pregnancy and normal pregnancy, the cardinal phenotype of dNK cells were CD56brightCD16-CD3-. in the pregnant women with Hypertensive disorder complicating pregnancy, the expression of intracellular cytokine IFN-γin dNK cells was significant lower than those in the normal pregnant women. It may suggests that the low IFN-γproduced by dNK cells is related with the bad placentation, which may be one of the pathogenesy of Hypertensive disorder complicating pregnancy.
