Objective:It is shown that tumor angiogenesis and ability of cancer cell migration play an important role in the growth progression and metastasis of Hepatocellular carcinoma(HCC). Tumor vessel forming could supply abundant nutrition and provide a metastasis for hepatic cancer cell.The forming of microvessel density(MVD) is as a of indexes which can reflect tumor angiogenesis in HCC.A mass of studies have shown the over expression of insulin-like growth factorⅡ(IGF-Ⅱ) in hepatocellular carcinoma tissues.It is a reseach aim that we will investgate the relationship between expression of insulin-like growth factorⅡand the forming of tumor microvessel density and discuss the significance both IGF-Ⅱand tumor microvessel forming in HCC occurring and evolution.Methods:Collected 40 cases of specimens of HCC tissue through the operated from 2006 to 2008 years as the first treated scheme,and enrolled all patients had not accepted radiation or chemical therapy or other anti-tumor treatment.S-P immunohistochemical method was applied to detecting the expression of IGF-Ⅱand MVD forming and distributing in hepatocarcinoma tissues.Results:1.The positive expression rate of IGF-Ⅱin HCC tissue was 85.0%(34/40),and overexpression of IGF-Ⅱin lower and moderate differentiated of HCC tissues was significantly higher than in high differentiated of HCC(P<0.05).The expressed level of IGF-Ⅱhad no correlatived with the clinical stages of the HCC(P>0.05).2.The HCC with small size from<5cm~5~10cm had MVD forming lower than HCC of larger ones(>10cm)(P<0.05).The HCC with low differentiation cell had higher MVD than the middle-high differentiated ones(P<0.05). The HCC in clinical stageⅡand stageⅢhad higher MVD than the cases in stageⅠ(P<0.05).It was revealed the positively relation that was the over-expression of IGF-Ⅱand MVD forming and distribution(r=0.727,P<0.01).Conclusion:The development and progression of hepatocellular carcinoma were related with the expression of IGF-Ⅱand MVD forming.The results suggested that IGF-Ⅱcould promote the forming of tumor microvessel in HCC.The high expression of IGF-Ⅱand the velocity of MVD forming could be reflect the malignant degree and used as one surveillance index for the prognosis and prediction of hepatocellular carcinoma.
