ObjectiveIn order to explore the mechanism of acupuncture effect on the cerebral infarction,we searched and identified the changed proteins of serum in the cerebral infarction patients before and alter electroacupuncture through two-dimensional electrophoresis(2-DE) and matrix-assisted laser desorption ionization time-of-fligh mass spectrometry(MALDI-TOF-MS) technologies, this will provide the experimental evidence for clinical treatment of cerebral infarction.MethodsSix abundant proteins were removed in serum samples by the Multiple Affinity Removal System Spin Cartridge Hu-6Hc(MARS).The serum proteins were separated by 2-DE.2-DE was scaned and acquire by Typhoon9200.The changed protein spots were selected by PDQuest software.The changed proteins were analyzed by MALDI-TOF-MS,and the mass spectrometric date were used to identify the protein through the general proteomics standard(GPS) and Mascot database.Western blotting was used to further confirm the expression of the changed proteins revealed by 2-DE.Results1.The protein expression profile of serum were established well in the cerebral infarction patients.2.Expression ratio of proteins was beyond 2.5±0.5 folds as the significant changed proteins.Expression of fifty serum proteins were significantly different between the pre-EA and post-EA.Among them,thirty-five proteins were down-regulated in the post-EA fifteen,proteins were up-regulated and compared with pre-EA.Nine significant changed protein spots were selected and identified by MALDI-TOF-MS.The down-regulate proteins are serpinG1,A1BG and APOA4;the up-regulated proteins are gelsolin,scavenger receptor(CFI),C3,C4b and APOH. Two protein spots were identified as the APOH.Western blotting analysis further confirmed expression of serpin G1 and gelsolin revealed by the 2-DE.ConclusionElectroacupuncture can increase immunity,inhibit cell apoptosis and promote functional recovery of nervous system through down-regulation expression of serpinG1,A1BG and APOA4, and up-regulation expressions of gelsolin,scavenger receptor(CFI),C3,C4b and APOH to The results suggested that the mechanism of electroacupuncture may be involved with multi-channed and multi-target protective effects on cerebral infarction.
