ObjectiveNeuroendocrine carcinomas(NEC) of the lung is the common maliganant tumor possessing endocrine function in lung cancer.It can be classified into four histopathological types:typical carcinoid tumor(TC),atypical carcinoid tumor (AC),large cell neuroendocrine carcinoma(LCNEC) and small cell lung carcinoma (SCLC).To provide more effective information for clinical practice and guiding treatment,the expression of following tumor-associated-gene proteins as CD133,LRP16,P53 and Ki67 were studied in NEC,and their correlations with the tumor biological characteristics and survival time of patients were analyzed further.MateriaIs and methodsThis study is a retrospective research.One hundred and seventy-nine cases of NEC of the lung from 1988 to 2008 in Chinese People's Liberation Army(PLA) General Hospital were reviewed.Cases of four types of NEC were selected according to the revised 1999 WHO Histological Typing of Lung and Pleural Tumors.Follow- up in 75 cases of patients' treatment and survival time were achieved through phone interview to reach a comprehensive clinical data.The immunohistochemical technique of non-biotin detection system and pressure cooker antigen retrievaling method were applied to detect the expression of CD133,LRP16,P53 and Ki67 proteins in paraffin embedded tissue.After that,the relationship between the expression of the four proteins and clinicopathological features as well as survival time was statistically analyzed.ResultsThe positive expression rates in expression of the four gene proteins were observed as follows:CD133 59%,LRP16 25%,P53 34%and Ki67 72%.Among them, CD133,LRP16,P53 and Ki67 were correlated with the typing of NEC(P<0.05),in which the expression of the four proteins was higher in SCLC than other three types.The expression of CD133,LRP16 and P53 was not related to clinical stage(P>0.05).The expression of CD133,LRP16,Ki67 and P53 showed significantly correlative with the prognosis of NEC by Log rank survival analysis(P<0.05).The positive expression of CD133,LRP16,Ki67 and P53 was corresponding to worse prognosis.The result of Cox survival analysis indicated that LRP16,P53,sex and TNM were independent prognostic factors for NEC(P<0.05).Conclusions1.CD133,LRP16,P53 and Ki67 may be related to the type of NEC;while CD133,LRP16,P53 and Ki67 may be related to clinical stage,but not to lymph node metastasis.2.CD133,LRP16,P53 and Ki67 may be significantly correlative with the prognosis of NEC.The positive expression of CD133,LRP16,P53 and Ki67 may indicate worse prognosis.3.LRP16,P53,sex and TNM were independent prognostic factors for NEC.
