[Objective] To study the effect of anti-adhesion isolated membranes of modified chitosan onsciatic nerve regeneration of rats and to testify the potential clinical application of modified chitosan in nerve repair.[Methods] 3 mm bilateral sciatic nerve was resected respectively in 45 SD rats, and the outermembrane was sutureed in tension. Package the suture of unilateral sciatic nerve randomly with anti-adhesion isolated membranes of modified chitosan, and establish bilateral control. 45 rats were randomly divided into three groups respectively, observe recovery time of claw reflection after operation, electromyogram and histological examinations were taken at 20days (group A),30days (group B),40 days (group C) postoperatively.[Results] Comparing bilateral pathological examination, the anti-adhesion isolatedmembranes of modified chitosan aggravated the inflammatory response of nerve stumps, With the degradation of the membranes, inflammation relieved gradually. the anti-adhesion isolated membranes of modified chitosan can relieve fiber hyperplasia of nerve tissue. 30 days after sciatic nerve injury, the speed of conduction velocity of regenerated nerve is faster in treatment side, density of nerve axonal is greater. There is a significance in the comparision of bilateral control in every points after 30 days of injury.[Conclusion] Anti-adhesion isolated membrane of modified chitosan aggravated theinflammatory response of peripheral nerve injury in early, but with the degradation of the membranes, inflammation relieved gradually. It could relieve fiber hyperplasia of nerve tissue and prevent adhesion, and it can improve recovery of conduction velocity and regeneration of nerve axon. Therefore anti-adhesion isolated membrane of modified chitosan could be used for the treatment of peripheral nerve injury.
